摘要
以小鼠体内金银花提取物绿原酸为研究对象,采用高效液相色谱法(high performance liquid chromatography,HPLC)测定小鼠血浆、肝脏、肾脏的绿原酸浓度并研究小鼠体内金银花提取物绿原酸的药代动力学。对小鼠灌胃金银花提取物绿原酸取血后,血浆样品用甲醇沉淀蛋白处理后经HPLC检测,用C18柱(4.6 mm×250 mm, 5μm)为色谱柱,以V (甲醇)︰V (0.2%磷酸溶液)=12︰88为流动相,柱温箱35℃,流速1.0 mL/min,检测波长327 nm,进样量20μL。结果表明:绿原酸的主峰保留时间为6.972 min,峰形正态分布,分离良好;绿原酸的线性方程为y=0.923 7x-1.012 9,相关系数R^(2)=0.999 4,绿原酸的进样浓度在3~28μg/mL范围内有良好线性关系;最低检测限和定量限分别为0.056 7μg/mL和0.248 8μg/mL,日内与日间精密度均小于15%,血浆、肝脏和肾脏的平均回收率为92.42%, 91.95%和89.86%,高、中、低3种浓度的水平回收率在0.33%~2.83%范围内,符合药代动力学研究要求。试验采用的HPLC法专属性好,适于进行绿原酸在小鼠体内的药代动力学研究。
The chlorogenic acid extracted from Flos Lonicerae in mice was used as the research object,the concentration of chlorogenic acid in plasma,liver and kidney of mice was determined by high performance liquid chromatography(HPLC),and the pharmacokinetics of chlorogenic acid in mice was studied.After oral administration of chlorogenic acid from Flos Lonicerae extract in mice,the plasma samples were precipitated with methanol and detected by HPLC,using a Cis column(4.6 mmx250 mm,5μm)as the chromatographic column,V(methanol):V(0.2%phosphoric acid solution)=12:88 as the mobile phase,column temperature 35℃,flow rate 1.0 mL/min,detection wavelength 327 nm,and injection volume 20μL.The results showed that the retention time of the main peak of chlorogenic acid was 6.972 min;The peak shape was normal distribution,and the separation was good;The linear equation of chlorogenic acid was y-0.9237x-1.0129;The correlation coefficient(R^(2))was 0.9994,and the injection concentration of chlorogenic acid had a good linear relationship in the range of 3-28μg/mL.The lowest detection limit and quantitative limit were 0.0567μg/mL and 0.2488μg/mL,respectively.The intra-day and inter-day precisions were less than 15%.The average recoveries of plasma,liver and kidney were 92.42%,91.95%and 89.86%,respectively.The horizontal recoveries of high,medium and low concentrations were in the range of 0.33%to 2.83%,which met the requirements of pharmacokinetic study.The HPLC method used in the experiment has good specificity and is suitable for the study of pharmacokinetics of chlorogenic acid in mice.
