低强度聚焦超声对新生小鼠大脑皮层发育及星形胶质细胞活化的影响

摘要 目的 观察低强度聚焦超声(LIFU)对新生小鼠大脑皮层发育及星形胶质细胞活化的影响。方法 将80只新生小鼠分为LIFU组(n=48)和假手术组(n=32)。对LIFU组小鼠以LIFU连续波辐照3天、每天5 min;将假手术组小鼠置入声场中,不施加声波。采用蛋白质印迹法检测皮层NFIX蛋白表达,以免疫荧光染色观察c-Fos、NFIX及胶质纤维酸性蛋白(GFAP)表达,以EdU染色观察皮层细胞增殖情况。结果 LIFU辐照后1.0、1.5、2.0、2.5及3.0 h, LIFU组小鼠大脑皮层c-Fos表达数量均较假手术组显著增加(P均<0.05),于1.5 h达峰,之后逐渐递减。辐照后1 h, LIFU组小鼠皮层NFIX蛋白表达量明显高于假手术组;辐照后1、2、3 h及1个月,LIFU组小鼠皮层NFIX数量均较假手术组明显增加(P均<0.05)。辐照后1个月,LIFU组小鼠大脑皮层GFAP阳性细胞数量明显增加,且细胞呈星形,从胞体发出大量长突起。辐照后16、24 h, LIFU组EdU阳性细胞数量较假手术组显著增多(P均<0.05),且增殖的EdU阳性细胞存在NFIX阳性细胞共表达。结论 LIFU可激活新生小鼠大脑皮层c-Fos表达,促进皮层中细胞增殖和GFAP星形胶质细胞增加,可能与NFIX有关。 Objective To observe the impact of low-intensity focused ultrasound(LIFU)on the development of cerebral cortex and the activation of astrocytes in newborn mice.Methods Totally 80 newborn mice were divided into LIFU group(n=48)and sham operation group(n=32).The mice in LIFU group were irradiated with LIFU continuous wave 5 min per day for 3 days,while those in sham operation group were placed in the sound field but without sound wave irradiation.Then the expression of NFIX protein in cortex was detected using Western blotting,the expression of c-Fos,NFIX and glial fibrillary acidic protein(GFAP)were observed with immunofluorescence staining,and the proliferation of cortical cells was observed using EdU staining.Results The expression of c-Fos in cortex of mice in LIFU group increased significantly at 1.0,1.5,2.0,2.5 and 3.0 h after irradiation in LIFU group compared with that in sham operated group(all P<0.05),which reached the highest value at 1.5 h and then decreased gradually.One hour after irradiation,the expression of NFIX protein in cortex in LIFU group was significantly higher than that in sham operation group,while 1,2,3 h and 1 month after irradiation,the number of NFIX in cortex in LIFU group increased significantly compared with that in sham operation group(all P<0.05).One month after LIFU irradiation,the number of GFAP positive cells in cortex in LIFU group increased significantly,which were star-shaped and with many long processes from the body.The number of EdU positive cells in LIFU group increased significantly compared with that in sham operation group at 16 and 24 h after HIFU irradiation(all P<0.05),and NFIX positive cells co-expressed in the proliferating EdU positive cells.Conclusion LIFU could activate the expression of c-Fos in cortex of newborn mice,promote the increase of GFAP astrocytes and the proliferation of cells in cortex,which might be related to NFIX.
