三叶青早花基因的克隆及其对光质与光周期信号的响应 被引量:1

Cloning of Early Flowering Genes in Tetrastigma hemsleyanum and Its Response to Light Quality and Photoperiod Signals

摘要 早花基因是植物生物钟感受光信号的重要分子元件,而生物钟与植物碳水化合物的代谢及运输有重要关系。为了解光质及光周期信号对三叶青的块根产量及早花基因表达的影响,本研究以怀玉山三叶青为试材,从三叶青的块根发育不同时期的转录组中筛选到3个差异表达的早花基因cDNA序列,通过RT-PCR法对其开放读码框进行扩增,并对其核酸序列及推导的蛋白质序列进行初步的生物信息学分析。测产实验表明,蓝光补充光照处理组产量最高,白光处理组产量最低,且与对照组呈显著差异;长日光周期可显著提高三叶青块根产量,而短日处理条件下块根产量显著降低。通过RT-qPCR方法分析表明红光显著促进叶片中早花基因的表达,红光处理及蓝光处理条件下,块根中早花基因的表达显著受到抑制;光周期信号对早花基因表达分析表明,3个早花基因的表达都受光周期调控,表现出不同的表达模式。 Early flowering genes are important component of plant circadian clocks in receiving light signals,and have an important role in plant carbohydrate metabolism and transportation.In order to analyze the influence of light quality and photoperiod signal on tuber yields and early flowering genes expression profile of Tetrastigma hemsleyanum Diels et Gilg,Huaiyushan T.hemsleyanum was used as the test material,and the open reading frames were amplified by RT-PCR based on three early flowering genes sequences that were differentially expressed in the T.hemsleyanum transcriptome at different development stage of tubers.Then bioinformatics analysis of the nuclear sequences and the predicted protein sequence of early flowering genes were performed.Studies ofRT-qPCR showed the red light could promote the expression of early flowering genes in leaves;under conditions of supplementing red and blue light,the expression of early flowering genes were inhibited.The analysis of yield showed that the blue light supplemental lighting had the highest yield,and the white supplemental lighting had the lowest yield.Both of them showed significantly different from the control group.The different photoperiods treatments demonstrates that LD condition could significantly increase the yield of root tuber and SD conditions reduced yields.The analysis of early-flowering gene expression profiles under different photoperiod conditions showed that the expression of the three early-flowering genes were all regulated by photoperiods and showed different expression patterns.
