基因治疗用于乳腺癌的研究现状

摘要 乳腺癌(breast cancer,BC)是女性最为常见的恶性肿瘤,也是全球女性癌症死亡的主要原因。手术、放疗和化疗等常规治疗的效果往往不如预期。基因治疗通过纠正缺陷基因和调节基因的表达来达到预期的治疗效果,目前已成为一种有前景的乳腺癌治疗策略。在乳腺癌的基因治疗中通常使用病毒载体和非病毒载体作为递送载体。病毒载体具有较高的转基因效率,但病毒载体存在免疫原性较高、具有插入突变的风险和生物安全性等问题。非病毒载体与病毒载体相比,细胞毒性、免疫原性和诱变性均较低。但它还需要解决基因转移效率较低、特异性不足、基因表达持续时间短等问题。本文综述了目前利用病毒载体和非病毒载体治疗乳腺癌的研究现状,这些基因治疗载体各有优缺点,因此本文对目前使用的基因治疗载体进行相关概述。 Breast cancer(BC)is the most common malignant tumor in women and the leading cause of cancer death in women worldwide.Conventional treatments like surgery,radiation and chemotherapy often don't work as well as expected.Gene therapy has become a promising strategy for breast cancer treatment by correcting defective genes and regulating gene expression to achieve the desired therapeutic effect.Viral and non-viral vectors are commonly used as delivery vectors in gene therapy of breast cancer.Viral vectors have high transgene efficiency,but there are some problems such as high immunogenicity,insertional mutation risk and biosafety of viral vectors.Non-viral vectors showed lower cytotoxicity,immunogenicity and mutagenicity than viral vectors.But it also needs to solve the low efficiency of gene transfer,lack of specificity,short duration of gene expression and other problems.This article reviews the current research status of the use of viral vectors and non-viral vectors in the treatment of breast cancer.These gene therapy vectors have their own advantages and disadvantages,so this article summarizes the current use of gene therapy vectors.
