治疗孤独症谱系障碍的组方用药规律及其核心中药组合作用机制研究 被引量:2

Study on the Prescription Patterns for Treatment of Autism Spectrum Disorders and Action Mechanism of Its Core Herbal Combinations

摘要 【目的】运用数据挖掘技术分析中药复方治疗孤独症谱系障碍(ASD)的组方用药规律,并运用网络药理学分析其核心中药组合治疗ASD的潜在作用机制。【方法】运用中医传承计算平台V3.0,对符合纳入标准的中药复方进行统计处理、关联分析、聚类分析;并运用网络药理学技术分析核心方的活性成分、靶点及对于ASD的作用通路。【结果】共筛选出中药处方97个,涉及168味中药,功效以补虚类多见;中药归经以脾经、心经居多;中药四气以温性最多;中药药味以甘味为主。可视化网络显示,白术、炙甘草、茯苓、石菖蒲、远志为核心中药。根据网络药理学预测出核心中药组合包含8-异戊烯基-山柰酚、常春藤皂苷元、3β-乙酰氧基苍术酮、DFV、甘草苷等多种主要活性成分,涉及AKT1、ESR1、TNF等关键靶点;其作用机制与胆碱能突触、靶点基因与神经活性配体-受体相互作用、癌症通路、化学致癌-受体活化通路、神经变性途径-多种疾病、多巴胺能突触等信号通路相关。【结论】中药复方治疗ASD以补虚为主,多从心经、脾经入手,其核心中药组合“白术-炙甘草-茯苓-石菖蒲-远志”可有效通过抗氧化、抗炎等作用调控与ASD相关的细胞信号通路,从而达到治疗的目的。 Objective To analyze the prescription patterns of Chinese herbal compound for the treatment of autism spectrum disorders(ASD)by data mining techniques,and to analyze the potential mechanism of action of its core herbal combinations for the treatment of ASD by network pharmacology. Methods The TCMICS V3.0 was used to perform statistical processing,correlation analysis and cluster analysis on the herbal compound prescriptions that met the inclusion criteria;and to analyze the active components,targets and pathways of action for ASD of the core prescriptions using network pharmacology techniques. Results A total of 97 herbal prescriptions were screened, involving 168 herbal medicines, the effects of which were mostly in the category of deficiencyreplenishment;meridian tropism of herbal medicines were mostly in the spleen and heart meridians;the four natures of herbal medicines were mostly warm nature;and the taste of herbal medicines was mainly sweet. The visualization network showed that Atractylodis Macrocephalae Rhizoma, Glycyrrhizae Preparata Radix, Poria,Acori Tatarinowii Rhizoma,Radix Polygalae were the core herbs. Based on the network pharmacology,the core herbal combination contained a variety of major active components such as 8-isopentenyl-kaempferol,hederagenin, 3β-acetoxycangenone, DFV and glycyrrhizin, involving key targets such as AKT1, ESR1 and TNF;the mechanism of action was related to signaling pathways such as cholinergic synapses,target gene and neuroactive ligand-receptor interactions, cancer pathways, chemical carcinogenesis-receptor activation pathways,neurodegenerative pathways-multiple diseases,and dopaminergic synapses. Conclusion The treatment of Chinese herbal compound for ASD is predominated by replenishing deficiency,and starts with heart meridian and spleen meridian. The core herbal combination“Atractylodis Macrocephalae Rhizoma,Glycyrrhizae Preparata Radix, Poria, Acori Tatarinowii Rhizoma, Radix Polygalae”can effectively regulate the cellular signaling pathways related to ASD thr
