摘要
目的 研究膀胱癌中免疫细胞的浸润模式,并探讨免疫细胞浸润与膀胱癌预后的关系.方法 从GEO数据库中下载膀胱癌的相关基因芯片数据GSE13507及相关临床数据,通过CIBERSORT软件利用反卷积法计算膀胱癌中22种免疫细胞的浸润情况,并利用R软件Sur-vival包进行生存分析,分析每种免疫细胞与膀胱癌生存预后的关系.结果 从GEO数据库下载GSE13507芯片中得到正常膀胱组织(68例)和膀胱癌组织(165例),采用反卷积法计算免疫细胞浸润比例,得到正常膀胱组织13例,膀胱癌组织15例,膀胱癌组织中浸润水平较高的免疫细胞为:M0巨噬细胞[(13.18±12.53)%]、M2巨噬细胞[(12.12±10.27)%]、活化的树突状细胞[(7.77±7.84)%]和调节性T细胞[(7.37±5.99)%].正常膀胱组织和膀胱癌组织的记忆性B细胞、滤泡辅助性T细胞、中性粒细胞浸润水平比较,差异有统计学意义(P=0.004、0.019、0.012).与膀胱癌具有相关性的免疫细胞包括:活化的肥大细胞与活化的树突状细胞(r=0.75)、中性粒细胞与嗜酸性粒细胞(r=0.72)、活化的树突状细胞与幼稚CD4 T细胞(r=0.68)、幼稚B细胞与滤泡辅助性T细胞(r=0.65)、静息态树突状细胞与未活化的CD4记忆性T细胞(r=0.64).膀胱癌中的免疫细胞浸润与预后具有相关性的包括:记忆性B细胞、幼稚B细胞、调节性T细胞(均P<0.05).结论 膀胱癌中存在不同程度的免疫细胞浸润,记忆性B细胞、幼稚B细胞、调节性T细胞可以成为膀胱癌生存预后的标志物和免疫治疗的靶点.
Objective To study the pattern of immune cell infiltration in bladder cancer and the relationship between immune cell infiltration and the prognosis of bladder cancer.Methods The bladder cancer related gene chip data GSE13507 and related clinical data were downloaded from the GEO database.The infiltration of 22 immune cells in bladder cancer was calculated by the CIBERSORT software using the convolution method,and the R software survival package was used for survival analysis to analyze the relationship between each immune cell and the survival prognosis of bladder cancer.Results Normal bladder tissues(68 cases)and bladder cancer tissues(165 cases)were obtained from CSE13507 chip downloaded from GEO database.The proportion of immune cell infiltration in bladder cancer was calculated by deconvolution method,and 13 normal bladder tissues and 15 bladder cancer tissues were obtained.The highly infiltrated immune cells in bladder cancer tissues were MO macrophages[(13.18±12.53)%],M2 macrophages[(12.12±10.27)%],activated dendritic cells[(7.77±7.84)%]and regulatory T cells[(7.37±5.99)%].There were significant differences in the infiltration levels of memory B cells,T follicular helper cells and neutrophils between normal bladder tissues and bladder cancer tissues(P=0.004,0.019,0.012).Immune cells associated with bladder cancer include:activated mast cells and activated dendritic cells(r=0.75),neutrophils and eosinophils(r=0.72),activated dendritic cells and naive CD4 T cells(r=0.68),naive B cells and follicular helper T cells(r=0.65),resting dendritic cells and unactivated CD4 memory T cells Cells(r=0.64).The infiltration of immune cells in bladder cancer was correlated with prognosis,including memory B cells,naive B cells and regulatory T cells(all P<0.05).Conclusions There are varying degrees of immune cell infiltration in bladder cancer,memory B cells,naive B cells,and regulatory T cells can be markers for survival and prognosis of bladder cancer and targets for immune therapy.
