摘要
【目的】文章旨在分析西门塔尔牛活体采卵-体外受精(OPU-IVF)体外胚胎生产效率的影响因素,为提高西门塔尔牛体外胚胎生产效率、优化西门塔尔牛体外胚胎生产流程提供生产依据。【方法】本实验以未经激素处理的西门塔尔牛卵丘卵母细胞复合体(COCs)、进口肉牛冻精为实验材料,通过卵母细胞活体采集(OPU)、体外成熟(IVM)、体外受精(IVF)、早期胚胎体外培养(IVC)等技术手段,对体外受精后第7 d囊胚率进行统计分析。【结果】不同公牛精液对囊胚发育有影响,不同供体母牛对囊胚发育率影响显著。【结论】相同卵母细胞利用率水平的供体母牛,回收COCs体外受精后,囊胚率与冻精有关(P<0.05);不同卵母细胞利用率水平的供体母牛,回收COCs体外受精后,囊胚率随卵母细胞利用率升高而升高(P<0.05)。
【Objective】The purpose of this paper is to analyze the factors affecting the in vitro embryo production efficiency of Simmental bovine OPU-IVF, so as to provide productive basis for improving the efficiency of beef cattle in vitro embryo production and optimizing the process of beef cattle in vitro embryo production.【Methods】In this experiment, the cumulus oocyte complex(COCs) of Simmental cattle without hormone treatment and the frozen semen of imported beef cattle were used as experimental materials.【Results】The blastocyst rate on the 7 day after in vitro fertilization was statistically analyzed by means of oocyte collection in vivo(OPU), in vitro maturation(IVM), in vitro fertilization(IVF) and early embryo culture(IVC).【Conclusion】The results showed that the blastocyst rate of donor cows with different oocyte utilization levels increased with the increase of oocyte utilization rate after in vitro fertilization of COCs, and the blastocyst rate of donor cows with the same oocyte utilization level was related to frozen semen.
