摘要
目的 荧光偏振试验(FPA)是一种新的试验方法,本文旨在对FPA检测布鲁氏菌病的可行性进行研究,以适应国际贸易对疫病检测方法发展的需要。方法 通过对FPA的分析敏感性、分析特异性、重复性、诊断敏感性和特异性、重现性的测试,评估FPA作为检测方法的符合性和有效性。结果 FPA的分析敏感性高于RBT、SAT、CFT,低于iELISA、cELISA;FPA的分析特异性高于RBT、SAT、iELISA,与CFT、cELISA相当;FPA的重复性测试结果为组内和组间的变异系数均小于20%;FPA的诊断特异性为99.1%(1119/1129),诊断敏感性为97%(34/35);FPA的重现性测试结果为10份测试样品实验室间的变异系数均小于20%。结论 本研究说明FPA用于布鲁氏菌病抗体检测是有效、可靠的,方法简单、快速、通量大。
Objective To study the validation of the fluorescence polarization assay(FPA) which is a new serological tests. Methods Evaluate the valid of FPA by the tests of analytical sensitivity,analytical specificity,repeatability,diagnostic sensitivity,diagnostic specificity. Results The analytical sensitivity of FPA is higher than RBT、SAT、CFT and lower than iELISA、cELISA.The analytical specificity of FPA is higher than RBT、SAT、cELISA and the same as CFT、i ELISA.The coefficient of variation interclass and between the lab are smaller than20%. The diagnostic sensitivity of FPA is 97%(34/35),the diagnostic specificity of FPA is 99.1%(1119/1129).Conclusion The study results suggest that the FPA for detection of antibody to Brucella abortus is valid and that the FPA is a very simple,very rapid,very reliable diagnostic method.
