摘要
目的探讨miR-211对程序性死亡-配体1(PD-L1)介导的肝癌细胞免疫逃逸作用及其相关机制。方法收集我院19例肝癌患者肿瘤组织,通过qRT-PCR检测肝癌组织中miR-211和PDL1表达水平并分析两者的相关性。检测人肝癌HepG2、HCCLM3细胞和人正常肝上皮细胞HL-02中miR-211和PD-L1表达水平。将HepG2细胞分为miR-NC组(转染miR-NC)、miR-211组(转染miR-211),检测2组细胞中miR-211和PD-L1表达水平以及NK-92细胞对肝癌细胞的杀伤率。采用荧光素酶报告基因实验检测miR-211和BIN1的靶向关系。再将HepG2细胞分为miR-NC+oe-NC组(转染miR-NC和oe-NC)、miR-211+oe-NC组(转染miR-211和oe-NC)及miR-211+oe-BIN1组(转染miR-211和oe-BIN1),检测各组细胞中miR-211、BIN1和PD-L1表达及NK-92细胞对肝癌细胞的杀伤率。结果肝癌患者肿瘤组织中miR-211和PD-L1表达呈正相关(r=0.6007,P=0.001)。与HL-02细胞比较,HepG2和HCCLM3细胞中miR-211、PD-L1表达水平升高(P<0.05);与miR-NC组比较,miR-211组miR-211、PD-L1表达水平升高(P<0.05),NK-92细胞对肝癌细胞的杀伤率降低(P<0.05)。miR-211靶向结合并抑制BIN1。与miR-NC+oe-NC组比较,miR-211+oe-NC组miR-211、PD-L1表达水平升高(P<0.05),BIN1表达水平降低(P<0.05),NK-92细胞对肝癌细胞的杀伤率降低(P<0.05)。与miR-211+oe-NC组比较,miR-211+oe-BIN1组BIN1表达水平升高(P<0.05),PD-L1表达水平降低(P<0.05),NK-92细胞对肝癌细胞的杀伤率升高(P<0.05)。结论在肝癌细胞中,miR-211靶向抑制BIN1表达,从而促进PD-L1依赖的免疫逃逸。
Objective To investigate the effect of miR-211 on programmed cell death 1 ligand 1(PD-L1)mediated immune escape of hepatocellular carcinoma cells and its related mechanism.Methods The tumor tissues of 19 hepatocellular carcinoma patients were collected,and the expression levels of miR-211 and PD-L1 in tumor tissues were detected by qRT-PCR,and their correlation was analyzed.The expression levels of miR-211 and PD-L1 in human hepatocellular carcinoma cells of HepG2 and HCCLM3,and human normal hepatic epithelial cells of HL-02 were detected.HepG2 cells were divided into the control group(transfected with miR-NC)and miR-211 group(transfected with miR-211),and the expression levels of miR-211 and PD-L1,and the cell killing rate of NK-92 cells to hepatocellular carcinoma cells in the two groups were detected.The targeting relationship between miR-211 and BIN1 was detected by the luciferase reporting assay.HepG2 cells were then divided into the miR-NC+oe-NC group(transfected with miR-NC and oe-NC),miR-211+oe-NC group(transfected with miR-211 and oe-NC),and miR-211+oe-BIN1 group(transfected with miR-211 and oe-BIN1),and the expression levels of miR-211,BIN1 and PD-L1 and the cell killing rate of NK-92 cells to hepatocellular carcinoma cells in each group were detected.Results The expression levels of miR-211 and PD-L1 in tumor tissues of hepatocellular carcinoma patients were positively correlated(r=0.6007,P=0.001).Compared with HL-02 cells,the expression levels of miR-211 and PD-L1 in HepG2 and HCCLM3 cells were increased(P<0.05).Compared with the miR-NC group,the expression levels of miR-211 and PD-L1 in the miR-211 group were increased(P<0.05),and the cell killing rate of NK-92 cells to the hepatocellular carcinoma cells was decreased(P<0.05).miR-211 targeted binding BIN1 and inhibited its expression.Compared with the miR-NC+oe-NC group,the expression levels of miR-211 and PD-L1 in the miR-211+oe-NC group were increased(P<0.05),the expression level of BIN1 was decreased(P<0.05),and the killing rate of NK-92 cells to
