摘要
以溶胶-凝胶法合成双模型介孔SiO_(2)材料,并以该材料为载体吸附固定化脂肪酶。通过BJH法孔径分析和XRD表征表明,制备的载体呈现出孔径约为3nm和30nm的硅基结构的特征。以考马斯亮蓝法考察固载率,当固定化时间为2h、缓冲溶液pH值为8、固载温度为40℃时,固定化脂肪酶的固载率最大,可达到72.9%。
The bimodel mesoporous silica was synthesized by sol-gel method,and lipase was adsorbed and immobilized on this material.The analysis by BJH method and XRD characterization showed that the prepared support exhibited the characteristics of silicon-based structure with pore size of about 3nm and 30nm.Bradford method was used to investigate the immobilization rate.When the immobilization time was 2h,the pH of the buffer solution was8,and the immobilization temperature was 40℃,the immobilization rate was the highest,reaching 72.9%.
