摘要
目的通过观察腺苷预处理的脑缺血再灌注大鼠脑损伤程度及白细胞介素(IL)1β、IL-6、环氧化酶2(COX-2)表达变化,探讨腺苷对脑缺血再灌注损伤脑组织的保护作用。方法将85只雄性SD大鼠随机分为空白组10只及假手术组、模型组、腺苷组各25只。模型组和腺苷组采用线栓法制备右侧大脑中动脉缺血再灌注模型,腺苷组造模前连续3 d腹腔注射腺苷(1.5 mg/kg)溶液2 mL、假手术组和模型组造模前注射等量生理盐水,假手术组术中不插入线栓,空白组不予任何处理。于再灌注后24 h进行神经功能缺损评分评估神经功能,进行TTC染色测算脑梗死体积,采用TUNEL染色观察脑细胞凋亡情况;分别于再灌注后6、12、24、48 h采用ELISA法检测脑组织中的IL-1β、IL-6,采用免疫组化法检测脑组织中的COX-2。结果腺苷组、模型组大鼠神经缺损评分、脑梗死体积占比及脑细胞凋亡率高于空白组和假手术组,腺苷组大鼠神经缺损评分、脑梗死体积占比及脑细胞凋亡率低于模型组(P均<0.05)。模型组、腺苷组大鼠脑组织中IL-1β、IL-6、COX-2表达高于空白组和假手术组,腺苷组IL-1β、IL-6、COX-2表达低于模型组(P均<0.05)。结论腺苷预处理能够减轻脑缺血再灌注后大鼠脑损伤,可能是通过调节IL-1β、IL-6、COX-2表达发挥神经保护作用。
Objective To observe the degree of brain injury and the changes of interleukin(IL)-1β,IL-6,and cy⁃clooxygenase 2(COX-2)levels in rats with cerebral ischemia-reperfusion injury after adenosine preconditioning and to in⁃vestigate the protective effect of adenosine on brain tissues.Methods Eighty-five male SD rats were randomly divided in⁃to the blank group(10 rats),sham operation group(25 rats),model group(25 rats),and adenosine group(25 rats).In the model group and adenosine group,we prepared the right middle cerebral artery ischemia reperfusion models by thread embolism method.The rats in the adenosine group were injected with 2 mL of adenosine(1.5 mg/kg)solution intraperito⁃neally for 3 consecutive days before modeling.The rats in the sham operation group were injected with the same amount of normal saline before modeling.In the sham operation group,we did not insert thread embolism during operation,and the rats in the blank group were not treated.At 24 h after reperfusion,the neurological function was evaluated by the neurologi⁃cal deficit score,the volume of cerebral infarction was measured by TTC staining,and the apoptosis of brain cells was ob⁃served by TUNEL staining;IL-1βand IL-6 in the brain tissues were detected by ELISA,and COX-2 in the brain tissues was detected by immunohistochemistry at 6,12,24 and 48 h after reperfusion.Results The nerve defect score,the pro⁃portion of cerebral infarction volume and the apoptosis rate of brain cells in the adenosine group and model group were high⁃er than those in the blank group and sham operation group,while the nerve defect score,the proportion of cerebral infarc⁃tion volume and the apoptosis rate of brain cells in the adenosine group were lower than those in the model group(all P<0.05).The expression levels of IL-1β,IL-6,and COX-2 in the brain tissues of rats in the model group and adenosine group were higher than those in the blank group and sham operation group,while the expression levels of IL-1β,IL-6 and COX-2 in the adenosine group wer
