腺苷A1受体阻断剂对电针预处理诱导脊髓缺血耐受作用的影响 被引量:6

Effects of adenosine A1 receptor antagonist on ischemic tolerance induced by repeated electroacupuncture preconditioning in rabbits with spinal cord ischemia

摘要 目的:探讨腺苷A1受体阻断剂对电针预处理诱导脊髓缺血耐受作用的影响。方法:40只雄性新西兰大白兔随机分成对照组、戊巴比妥钠组、二甲基亚砜(DMSO)组、电针(EA)加8环戊基1,3二丙基黄嘌呤(EA+DPCPX)组和EA预处理组(n=8)。各组给予相应药物和(或)电针预处理,连续5d。最后一次处理后24h,阻闭肾下腹主动脉20min,制作兔脊髓缺血模型;再灌注后4、8、12、24和48h分别进行动物后肢运动神经功能评分。再灌注48h后处死动物,取脊髓(L57),石蜡包埋、切片,行组织病理学观察。结果:EA预处理组各时间点运动神经功能评分及脊髓前角正常运动神经细胞数明显高于EA+DPCPX组(P均<0.01),EA+DPCPX组与对照组比较差异有显著性(P均<0.05);对照组、DMSO组与戊巴比妥钠组各时间点运动神经功能评分及脊髓前角运动神经元计数差异均无显著性(P均>0.05)。结论:重复电针预处理诱导脊髓缺血耐受的机制是通过激活腺苷A1受体而实现的,腺苷A1受体阻断剂能部分拮抗电针抗脊髓缺血再灌注损伤的预处理效应,提示电针预处理效应是多因素综合作用的结果。 Objective: To investigate the effect of adenosine A1 receptor antagonist on ischemic tolerance induced by repeated electroacupuncture (EA) preconditioning in rabbits with spinal cord ischemia.Methods: Forty New Zealand white rabbits were randomly divided into five groups (n=8). Animals in the control (C) group, sodium pentobarbital (SP) group and dimethyl sulfoxide (DMSO) group received normal saline, SP and DMSO respectively each day for 5 days. Animals in EA group were anesthetized with SP and received EA at the Weizhong acupoint (委中穴) 1 hour each day for 5 days ,animals in EA+8-cyclopentyl-1,3-dipropylxanthine (DPCPX) group received DPCPX 30 minutes before preconditioning additionally.Twenty-four hours after last treatment, spinal cord ischemia model was replicated by infarenal aortic occlusion for 20 minutes. Hind -limb motor neurological function was determined at 4, 8, 12, 24 and 48 hours after reperfusion, and the histopathologic changes of spinal cords (L5 - 7) were observed at 48 hours after reperfusion. Results: The hind -limb motor neurological function and normal neurons of anterior horn in spinal cord of the EA group were significantly higher than those of the EA+DPCPX group (both P〈0.01),while those of the EA+DPCPX group were significantly higher than those of the control group (both P〈0.05). No significant difference on the above -mentioned parameters was observed among the control group, DMSO group and SP group at each time point (all P〉0.05). Conclusion: The ischemic tolerance induced by repeated EA is possibly via the activation of adenosine Al receptor in spinal cord. Adenosine Al receptor antagonist has the effect of partially antagonising preconditioning effect of EA on anti - ischemia/reperfusion injury in spinal cord, which implies that the effect is the result of multi -factors.
