紫草油对激素依赖性皮炎豚鼠模型的影响及机制 被引量:2

The Effect and Mechanism of Lithospermum Oil on Hormone-Dependent Dermatitis in Guinea Pig Model

摘要 目的探究紫草油对激素依赖性皮炎(HDD)豚鼠模型的影响及机制。方法采用完全随机方法将32只豚鼠分为空白对照组、模型组、他克莫司组、紫草油组。除空白对照组外,其余组豚鼠采用0.5%丙酸氯倍他索酊剂建立HDD模型,模型建立后,模型组外涂生理盐水,各给药组外涂相应药物,连续3周。ELISA法测定血清IFN-γ、IL-4、IgE水平;免疫组织化学和Western blot法检测表皮丝聚蛋白(FLG)、兜甲蛋白(LOR)及半脱氨酸蛋白酶-14(Caspase-14)表达情况。结果较之于空白组,模型组豚鼠血清IFN-γ、IL-4、IgE水平升高[(8.25±1.39)pg/mL vs.(14.78±1.47)pg/mL、(7.95±0.45)pg/mL vs.(10.38±0.39)pg/mL、(43.37±5.81)μg/mL vs.(60.20±3.26)μg/mL,P<0.05],表皮FLG、LOR和Caspase-14表达水平降低,差异均有统计学意义(P<0.05)。较之于模型组,紫草油组及他克莫司组豚鼠血清IFN-γ、IL-4、IgE水平降低[(14.78±1.47)pg/mL、(9.13±1.45)pg/mL、(10.43±1.92)pg/mL;(10.38±0.39)pg/mL、(8.07±0.39)pg/mL、(7.84±0.47)pg/mL;(60.20±3.26)μg/mL、(46.40±6.56)μg/mL、(41.71±3.25)μg/mL,P<0.05],表皮FLG、LOR和Caspase-14表达水平升高,差异均有统计学意义(P<0.05)。结论紫草油对豚鼠HDD具有改善作用,其机制可能与抑制皮肤炎症反应、促进表皮皮肤屏障形成有关。 Objective This study was designed to investigate the effects and mechanism of lithospermum oil on hormone-dependent dermatitis(HDD) in guinea pigs.Methods Totally, 32 male guinea pigs were recruited and randomly divided into normal group, model group, tacrolimus ointment group, lithospermum oil group.The HDD guinea pigs model was established by the 0.5% Clobetasol propionate tincture.After HDD model establishment, model group smeared normal saline, the rest of the group with corresponding drugs for three weeks.The contents of IFN-γ,IL-4 and IgE in the guinea pigs serum were detected by the ELISA;the protein expression levels of FLG,LOR and Caspase-14 in the epidermis of guinea pigs were detected by the immunohistochemical and Western blot method.Results Compared with the normal group, the IFN-γ,IL-4 and IgE of guinea pigs serum in the model group were significantly increased[(8.25±1.39)pg/mL vs.(14.78±1.47)pg/mL,(7.95±0.45) pg/mL vs.(10.38±0.39)pg/mL,(43.37±5.81) μg/mL vs.(60.20±3.26)μg/mL;P<0.05];the FLG,LOR and Caspase-14 of guinea pigs epidermis in the model group were significantly decreased(P<0.05). Compared with the model group, the IFN-γ,IL-4 and IgE of guinea pigs serum in the lithospermum oil group and tacrolimus ointment group were significantly decreased[(14.78±1.47)pg/mL,(9.13±1.45)pg/mL,(10.43±1.92)pg/mL;(10.38±0.39)pg/mL,(8.07±0.39)pg/mL,(7.84±0.47)pg/mL;(60.20±3.26)μg/mL,(46.40±6.56)μg/mL,(41.71±3.25)μg/mL,P<0.05];the FLG,LOR and Caspase-14 of guinea pigs epidermis in the tacrolimus ointment group and lithospermum oil group were significantly increased(P<0.05).Conclusion Lithospermum oil can improve guinea pig HDD, and the mechanism may be related to inhibiting skin inflammation and promoting the formation of epidermal skin barrier.
