摘要
目的建立莲须的超高效液相色谱法(UPLC)指纹图谱,并建立其异槲皮苷的含量测定方法。方法色谱柱为Waters Acquity UPLC HSS T3,1.8μm,2.1×100 mm;以乙腈(A)-0.2%乙酸(B)为流动相进行梯度洗脱;流速0.2 mL·min^(-1);柱温:30℃;进样量3μL;检测波长300 nm。建立莲须药材的指纹图谱和异槲皮苷含量分析方法,并对19批不同产地莲须药材进行测定。结果指纹图谱及含量测定方法的各项方法学验证均良好,19批莲须与对照指纹图谱的相似度在0.883~0.995,标定了7个共有峰。主要成分异槲皮苷线性关系良好(R^(2)>0.999),加样回收率在91.58%~99.70%,RSD为3.54%,19批莲须异槲皮苷含量在0.011%~0.033%。结论所建立的莲须UPLC指纹图谱及异槲皮苷含量分析方法专属性强、分离度好、灵敏度高,可为莲须药材的质量控制与评价提供参考。
OBJECTIVE To establish the ultraperformance liquid chromatography(UPLC)fingerprint of Nelumbinis stamen,and to establish a quantitative analysis method for isoquercitrin of Nelumbinis stamen.METHODS WATERS ACQUITY UPLC HSS T3(100 mm×2.1 mm,1.8μm)column was used with mobile phase of acetonitrile(A)-0.1%acetic acid solution(B).The flow rate was 0.2 mL·min^(-1).The detection wavelength was 300 nm.The injection volume was 3μL and the column temperature was 30℃.UPLC fingerprints of Nelumbinis stamen and a quantitative analysis method for isoquercitrin were established,and 19 batches of Nelumbinis stamen from different producing areas were determined.RESULTS The method validation of fingerprint and determination method was good,the similarities between 19 batches of Nelumbinis stamen substance benchmark and their control fingerprint were 0.883~0.995,7 common peaks were identified.The linear relationship of isoquercitrin was good in a certain range of concentration(R^(2)>0.999),the average recovery was 91.58%~99.70%(RSD 3.54%),and the contents of isoquercitrin was 0.011%~0.033%.CONCLUSION The established UPLC fingerprint and content determination methods have strong specificity,good resolution and high sensitivity,it can provide reference for the quality control and evaluation of Nelumbinis stamen.
