摘要
目的:研究婴幼儿配方奶粉中黄曲霉毒素M_(1)的超高效液相色谱测定方法。方法:样品经甲醇-水溶液提取,上清液稀释,经免疫亲和柱净化和富集,再氮吹复溶后,用超高效液相色谱仪(荧光检测器)分析测定。结果:黄曲霉毒素M_(1)在0.0515~5.1500 ng·mL^(-1)线性良好,回归方程为Y=4.00×10^(5)X-8.74×10^(3),相关系数为0.999867,检出限为0.02μg·kg^(-1),定量限为0.05μg·kg^(-1);加标回收率在81.0%~90.3%,变异系数为0.72%~3.18%,且正确度较好。结论:该方法灵敏度高、准确性好,适用于测定婴幼儿配方奶粉中黄曲霉毒素M_(1)的测定。
Objective:To study the determination of aflatoxin M_(1)in infant formula milk powder by ultra-high performance liquid chromatography.Method:The sample was extracted by methanol-water solution,diluted by supernatant,purified and enriched by immunoaffinity column,and then dissolved by nitrogen blowing,then analyzed and determined by ultra-high performance liquid chromatography(fluorescence detector).Result:The linearity of aflatoxin M_(1)was good in 0.0515~5.1500 ng·mL^(-1),the regression equation was Y=4.00×10^(5)X-8.74×10^(3),the correlation coefficient was 0.999867,and the detection limit was 0.02μg·kg^(-1),limit of quantitation was 0.05μg·kg^(-1);The recovery rate of spiking was 81.0%~90.3%,CV was 0.72%~3.18%,and the accuracy was good.Conclusion:The method has high sensitivity and accuracy,and is suitable for the determination of aflatoxin M_(1)in infant formula milk powder.
