宫颈癌高危型HPV感染的影响因素与ERK1/2信号通路关键因子 被引量:5

Influencing factors of high-risk HPV infection in cervical cancer and key factors of ERK1/2 signaling pathway

摘要 目的 分析宫颈癌高危型人乳头状瘤病毒(hrHPV)感染的影响因素与细胞外信号调节激酶1/2(ERK1/2)信号通路关键因子.方法 选择2015年3月-2021年3月于湖北省妇幼保健院和襄阳市中心医院进行宫颈hrHPV DNA筛查检查且同时进行宫颈活检的191例患者,收集患者宫颈上皮内瘤变组织标本90份(宫颈上皮内瘤变组)和宫颈癌组织标本101份(宫颈癌组),另收集同期于医院进行子宫全切术的慢性宫颈炎组织标本60份(慢性宫颈炎组),采用第二代杂交捕获法(HC-Ⅱ)检测患者hrHPV感染情况,采用免疫组化法检测ERK1/2信号通路关键因子蛋白表达情况.结果 宫颈癌组hrHPV感染阳性率高于宫颈上皮内瘤变组和慢性宫颈炎组(P<0.05);宫颈癌组p-ERK1/2、p-c-Fos阳性率高于宫颈上皮内瘤变组和慢性宫颈炎组(P<0.05),p-c-Fun阳性率高于慢性宫颈炎组(P<0.05);低分化、Ⅱ+Ⅲ期以及有淋巴结转移宫颈癌患者组织中p-ERK1/2、p-c-Fos和p-c-Fun阳性率高于中高分化、Ⅰ期以及无淋巴结转移患者(P<0.05);多因素Logistic回归分析,结果显示,p-ERK1/2、p-c-Fos、p-c-Fun阳性是宫颈癌hrHPV感染的影响因素(P<0.05).结论 宫颈癌中ERK1/2信号通路关键因子明显上调表达,且ERK1/2信号通路关键因子上调表达与hrHPV感染密切相关,可能发挥共同促进宫颈癌发生发展的作用. OBJECTIVE To analyze the influencing factors of high-risk human papillomavirus(hrHPV)infection in cervical cancer and the key factors of extracellular signal-regulated kinases 1 and 2(ERK1/2)signaling pathway.METHODS Totally 191 patients who underwent cervical hrHPV DNA screening and cervical biopsy in Hubei Provincial Maternal and Child Health Hospital and Xiangyang Central Hospital between Mar.2015 and Mar.2021 were selected,and 90 cervical intraepithelial neoplasia tissue specimens(cervical intraepithelial neoplasia group)and 10l cervical cancer tissue specimens(cervical cancer group)were collected.Another 60 specimens of chronic cervicitis tissues(chronic cervicitis group)were collected from patients who underwent total hysterectomy during the same period in the hospital were collected.The second-generation hybrid capture(HC-Ⅱ)was used to detect hrHPV infection in patients,and immunohistochemistry was applied to detect the protein expressions of key factors of ERK1/2 signaling pathway.RESULTS The positive rate of hrHPV infection in cervical cancer group was higher than that in cervical intraepithelial neoplasia group and chronic cervicitis group(P<0.05).The positive rates of p-ERK1/2 and p-c-Fos were higher in cervical cancer group than those in cervical intraepithelial neoplasia group and chronic cervicitis group(P<0.05),and the positive rate of p-c-Fun was higher than that in chronic cervicitis group(P<0.05).The positive rates of p-ERK1/2,p-c-Fos and p-c-Fun in tissues of patients with low differentiation,stage II+III and lymph node metastasis were higher than those of patients with moderate-to-high differentiation,stage I and absence of lymph node metastasis(P<0.05).Multivariate Logistic regression analysis showed that the positive expressions of p-ERK1/2,p-c-Fos and p-c-Fun were the influencing factors of hrHPV infection in cervical cancer(P<0.05).CONCLUSION The key factors of ERK1/2 signaling pathway were significantly up-regulated in cervical cancer,and the up-regulations of key factors of ERK1/2 sign
