摘要
目的:了解黄芪多糖(astragalus polysaccharides,APS)作用后的人结直肠癌(colorectal cancer,CRC)细胞中血管内皮生长因子C(vascular endothelial growth factor-C,VEGF-C)的表达,探讨其对CRC淋巴结转移的影响及可能机制。方法:对不同浓度APS处理后的LoVo细胞进行划痕实验,并应用Western印迹法和RT-PCR法分析VEGF-C/LYVE-1的表达。结果:与对照组相比,APS作用后的LoVo细胞中VEGF-C和LYVE-1的蛋白表达降低,且随浓度梯度降低(P<0.05)。与对照组相比,APS作用后的LoVo细胞中VEGF-C和LYVE-1基因的相对荧光表达量随浓度梯度降低,差异均有统计学意义(P<0.05)。结论:APS可能参与肿瘤淋巴管的生成,在CRC淋巴转移中发挥重要作用,且APS可能是一种潜在的肿瘤淋巴管生成的抗淋巴管生成剂,可用于预防和治疗CRC淋巴转移。
Objective:To survey the expressions of VEGF-C in human colorectal cancer cells after the actions of APS,and explore its effects on lymph node metastasis in colorectal cancer and the possible mechanism.Methods:Scratch experiment was performed on LoVo cells treated with different concentrations of APS,Western blot and RT-PCR method were applied to analyze the expressions of VEGF-C/LYVE-1.Results:Compared with the control group,the expressions of VEGF-C and LYVE-1 protein reduced in LoVo cells after the actions of APS,and the protein relative expressions lowered when APS concentration reduced(P<0.05).Compared with the control group,the relative fluorescence expressions of VEGF-C and LYVE-1 genes in LoVo cells lowered after the actions of APS with the concentration gradient,and the difference had statistical meaning(P<0.05).Conclusion:APS might be involved in tumor lymphangiogenesis and play an important role in lymph node metastasis,and APS might be a potential anti-lymphangiogenic agent of tumor lymphangiogenesis,which could be used to prevent and treat lymphatic metastasis of colorectal cancer.
