丁酸梭菌对断奶仔猪盲肠菌群结构和产丁酸代谢的影响 被引量:2

摘要 为探究丁酸梭菌对断奶仔猪盲肠菌群结构和产丁酸代谢的影响,试验选取162只日龄为(25±1)d的“杜×长×大”三元杂交断奶仔猪,随机分成3组,对照组(Ctrl组)给予常规饲料喂养,试验组在对照组的基础上分别加入浓度为500 mg/kg(CB500组)和2000 mg/kg(CB2000组)的丁酸梭菌,试验周期为30 d。试验完成后,采用高通量测序技术和实时荧光定量(RT-qPCR)对盲肠菌群结构、产丁酸的功能基因丁酸激酶和丁酰-CoA:乙酰-CoA的表达水平进行测定分析,借助于气相色谱法对仔猪体内短链脂肪酸的含量进行测定分析。试验结果显示:(1)在门水平,仔猪盲肠优势菌门为厚壁菌门(Firmicutes)、变形菌门(Proteobacteria)、拟杆菌门(Bacteroidetes)和放线菌门(Actinobacteria);相比Ctrl组而言,CB2000组的变形菌门和广古菌门(Euryarchaeota)相对丰度分别下降了1.93%和6.62%(P<0.05)。在属水平,相比Ctrl组而言,试验组土孢杆菌属(Terrisporobacter)、库特氏菌属(Kurthia)和链球菌属(Streptococcus)的相对丰度分别升高了0.38%、0.71%和0.03%(P<0.05);弯曲菌属(Campylobacter)和瘤胃菌属(Ruminococcaceae_UCG-014)的相对丰度分别下降了5.5%和0.72%(P<0.05)。(2)丁酸梭菌处理使断奶仔猪盲肠中丁酸、戊酸和总短链脂肪酸等含量增多,其中丁酸含量增加趋势较为明显(P=0.064),特别是CB2000组丁酸含量比对照组增加了28.0%。(3)与Ctrl组相比,丁酸梭菌处理组的丁酸激酶和丁酰-CoA:乙酰-CoA转移酶的相对表达含量增多,尤其是丁酰-CoA:乙酰-CoA转移酶的表达量增加趋势较为显著(P<0.05),且CB2000组丁酰-CoA:乙酰-CoA转移酶的表达量与Ctrl组相比增加了15%。由此说明,丁酸梭菌处理能够改善仔猪盲肠菌群结构,增加菌群代谢产生的短链脂肪酸含量,从而更好地改善肠道内环境的稳态,有利于断奶仔猪的健康生长。 This paper focused on the specific effects of Clostridium butyricum on the microbial biological structure and metabolic process in the cecum of piglets who had interrupted breastfeeding after birth.162 Duroc land Yorkshire piglets with a birth time of(26±1)days were selected as the research objects,and they were randomly divided into three different experimental groups:the control group(Ctrl group)was fed with conventional feed for the basic day,CB500 group,and CB2000 group,which were fed with the basic diet supplemented with 500 mg/kg and 2000 mg/kg of Clostridium butyricum,respectively.The piglets had been raised for 30 days in this study.At day 30 of this experiment,the cecal content was collected for the analysis of microbiota structure and the determination of short chain fatty acid contents by high-throughput sequencing and gas chromatography,respectively while the segments of cecum was harvested for the measurement of the relative gene expression of butyryl-Co A:acetyl-Co A With the help of real-time quantitative PCR technology,the biological genes such as acetic acid helper enzyme and butyrate kinase were determined and analyzed.The experimental results showed that:(1)At the phylum level,the dominant flora mainly included Firmicutes,Proteus and actinomycetes;Compared with the ctrl group,the abundance of Proteobacteria and Euryarchaeota in CB2000 group decreased by 1.34%and 6.62%,respectively(P<0.05).At the genus level,relative abundance Terrisporobacter,Kurthia,Streptococcus was significantly increased by 0.38%,0.71%and 0.03%(P<0.05);The relative abundance of conditional pathogens such as Campylobacter and Ruminococcaceae_UCG-014 was significantly decreased by 5.5%and 0.72%,respectively(P<0.05).(2)The treatment with Clostridium butyricum increased the contents of butyric acid,isobutyric acid,valeric acid,isovaleric acid,and TSCFAs in the cecum of weaned piglets,where the content of butyric acid in the CB2000 groups gave a significant increase by 28.0%compared to that in the Ctrl group.(3)The relative gen
