藏药波棱瓜花多糖抗氧化活性研究

摘要 本研究旨在探讨藏药波棱瓜花多糖的抗氧化活性。试验设抗坏血酸(VC)对照组和波棱瓜花多糖组,通过测定DPPH·自由基清除率、Fe^(3+)还原力、·OH自由基清除率、ABTS·^(+)自由基清除率,以及S180荷瘤小鼠大脑、心脏、肝脏和血液中总超氧化物歧化酶(T-SOD)活性、过氧化氢酶(CAT)活性、丙二醛(MDA)含量,比较对照组和波棱瓜花多糖组抗氧化活性。结果显示,与对照组相比较,藏药波棱瓜花多糖组清除ABTS·^(+)、DPPH·、·OH等自由基的活性较弱,对Fe^(3+)离子还原能力不明显,且在0.1~1.0 mg/mL质量浓度范围内,其清除力和还原能力表现为浓度依赖性;多糖组能提高荷瘤小鼠血清中CAT活性,同时也可降低荷瘤小鼠血液高MDA含量;多糖组可恢复荷瘤小鼠血液低下的T-SOD活力,高剂量(200 mg/mL)多糖组可降低荷瘤小鼠大脑亢进的T-SOD活力至健康水平。结果表明,波棱瓜花多糖对ABTS·^(+)、DPPH·、·OH等自由基的清除活性不及VC,但升高荷瘤小鼠血清中低下的CAT活性和降低亢进的MDA含量的作用优于VC,对T-SOD活性的影响表现为“机能状态双向性”。 The purpose of this study was to explore the antioxidant effect of Tibetan medicine Herpetospermi Pedunculosi Flos polysaccharide. The study was divided into vitamin C control group and polysaccharide group. By measuring DPPH·radical scavenging rate, Fe^(3+)reducing power, ·OH radical scavenging rate, ABTS·^(+)radical scavenging rate, total superoxide dismutase(T-SOD) activity, catalase(CAT) activity and malondialdehyde(MDA) content in brain, heart, liver and blood of S180 tumor-burdened mice, the antioxidant activity of the control group and Herpetospermi Pedunculosi Flos polysaccharide was compared. The results showed that compared with the control group, Herpetospermi Pedunculosi Flos polysaccharide had lower ability of scavenging ABTS·^(+), DPPH· and ·OH, had no obvious effect on Fe^(3+)ion reduction, meanwhile its removal and reduction ability present a concentration dependence relationship at 0.1-1.0 mg·mL-1. Herpetospermi Pedunculosi Flos polysaccharide could increase the low CAT activity of tumor-bearing mice. At the same time, it could also reduce the high blood MDA content of tumor-bearing mice, which was better than VC, and the polysaccharide could restore the low blood T-SOD activity of tumor-bearing mice, and the high concentration(200 mg·mL^(-1)) could reduce the brain hyperactive T-SOD activity of tumor-bearing mice to a healthy level. The results showed that polysaccharide had certain scavenging ability on ABTS·^(+), DPPH· and ·OH, and played a certain role in increasing the activity of CAT and decreasing the content of MDA in the blood of tumor-bearing mice. Its effect on the activity of T-SOD was “functional bidirectionality”.
