摘要
针对单一植物精油抗菌活性弱、易挥发、不稳定等弊端,研究制备出一种复合精油纳米乳,并对其进行质量评价。采用微量肉汤稀释法测定8种植物精油对大肠杆菌的最小抑菌浓度(MIC),并选取抗菌活性强的3种植物精油,采用棋盘法测定联合用药后的部分抑菌浓度指数(FIC),根据棋盘试验结果确定最佳复配处方,通过伪三元相图筛选植物精油纳米乳最佳处方,利用相转化法制备其纳米乳并进行质量评价。结果显示,成功制备复合植物精油纳米乳,其结构类型为水包油型,马尔文激光粒度仪测得纳米乳平均粒径(Z-average)为32.49 nm,多分散系数(PDI)为0.203,在高速离心试验、光照及温度稳定性试验中,制备的复合植物精油纳米乳状态均无明显变化。将复合植物精油纳米乳用常水稀释10~120倍,24 h后观察,溶液仍澄清透明,证明以不同比例混溶于饮水后,稳定性良好。复合植物精油纳米乳制备工艺简单,状态稳定,可用于畜禽混饮用药。
Because of weak antibacterial, volatile, instability and other problems of single plant essential oil, a compound essential oil nanoemulsion was prepared and its quality was evaluated. The minimum inhibitory concentration(MIC) of eight plant essential oils against Escherichia coli were determined by the micro broth dilution method. According to the results of checkerboard test, three essential oils with strong antibacterial activity were selected and the fractional inhibitory concentration indices(FIC) of their combination was determined, then the optimal formulation of compound essential oil nanoemulsion was screened by pseudo ternary phase diagram. Furthermore, the phase transformation method was used to prepare nanoemulsion and its quality evaluation was carried out. The results indicated that compound plant essential oil nanoemulsion was successfully prepared and its structure type is oil-in-water type. The z-average particle size was 32.49 nm and the polydispersion coefficient(PDI) was 0.203 by Malvin laser particle size analyzer. In the high speed centrifugation test, light stability test and temperature stability test, the state of the prepared nanoemulsion of compound essential oil still had no obvious change. The compound plant essential oil nanoemulsion was diluted 10-120 times in normal water for 24 h, and the nanoemulsion was still clear and transparent. It was proved that the compound plant essential oil nanoemulsion had good stability after being mixed in drinking water at different proportions. The preparation process of compound plant essential oil nanoemulsion is simple and the state of nano-emulsion is stable. It can be used via the drinking water administration in livestock and poultry, and has great application potential in the prevention and control of bacterial diseases of livestock and poultry.
