摘要
通过绘制伪三元相图筛选百里香酚纳米乳处方,采用透射电子显微镜(TEM)、激光粒度测定仪分别考察其形态和粒径,利用染色法鉴别纳米乳的类型,通过光照试验、高速离心试验、加速试验以及长期试验考察百里香酚纳米乳的稳定性。结果表明,百里香酚纳米乳的最佳制备工艺是百里香酚和乙酸乙酯(质量比1∶1)为油相,蒸馏水为水相,混合表面活性剂中吐温-80和无水乙醇的质量比(Km)为3∶1,混合表面活性剂与油相的质量比为9∶1。百里香酚纳米乳为水包油型。在透射电镜下呈圆球形,无粘连。其平均粒径为16.4nm,多分散系数(PDI)为0.062。在光照试验、高速离心试验、加速试验和长期试验中,百里香酚纳米乳均保持澄清透明,药物含量基本未发生变化。该制剂制备工艺简单,有优良的稳定性。
The optimal formulation of thymol nanoemulsion was screened by studying the pseudoternary phase diagrams.The drop shape and average droplet size of thymol nanoemulsion were measured by transmission electron microscope(TEM) and the laser granularity analysis instrument,respectively.Its type was determined through staining method.And its stability was inspected by light test,high speed centrifugalization test,acceleration test and long-term test.The results showed that the best prescription of thymol nanoemulsion was as follows:thymol and ethyl acetate(mass ratio:1∶1) as mixed oil phase,distilled water as water phase,Tween-80 and ethanol(Km=3∶1) as mixed surfactants,in which the mass ratio of mixed surfactants and mixed oil phase was 9∶1.Thymol Nanoemulsion was of oil-in-water.The TEM showed that drops of thymol nanoemulsion were spheroidal and without adhesion to each other.The average droplet size was 16.4 nm with PDI at 0.062.The Thymol Nanoemulsion was still clarity after light test,high speed centrifugalization test,acceleration test and long-term test.And the drug content was unchanged.The tests showed a fine stability of the thymol nanoemulsion.
