摘要
目的观察阳和汤对人三阴性乳腺癌细胞BT549凋亡及STAT1/p53信号通路的影响,探讨其抑制三阴性乳腺癌发展的可能机制。方法将BT549细胞分为空白对照组、阳性药组及阳和汤低、中、高剂量组,分别用灌胃蒸馏水、5-氟尿嘧啶及阳和汤低、中、高浓度药液后的血清进行干预,DAPI染色和流式细胞术检测细胞周期及凋亡,q RT-PCR检测细胞信号传导和转录激活因子(STAT)1、p53 mRNA表达,Western blot检测STAT1/p53信号通路相关蛋白STAT1、STAT3、p53及Bax蛋白表达。结果与空白对照组比较,阳和汤低、中、高剂量组细胞凋亡率明显升高(P<0.01),G0/G1期细胞比例增加,G2/M期细胞比例降低(P<0.05,P<0.01),阳和汤高剂量组细胞STAT1、p53 mRNA表达明显升高(P<0.01),阳和汤中、高剂量组细胞STAT1、p53、Bax蛋白表达明显升高(P<0.05,P<0.01),阳和汤低、中、高剂量组细胞STAT3蛋白表达明显降低(P<0.01)。结论阳和汤能诱导BT549细胞凋亡,其机制与调控STAT1/p53信号通路相关因子STAT1、STAT3、p53和Bax表达有关。
Objective To observe the effects of Yanghe Decoction on apoptosis and STAT1/p53 signaling pathway in triple-negative breast cancer cells BT549;To explore its possible mechanism of inhibiting the development of triple-negative breast cancer.Methods BT549 cells were divided into blank control group,positive drug group and Yanghe Decoction low-,medium-and high-dosage groups.Interventions were performed with the serum after gavage of distilled water,5-fluorouracil,and low-,medium-and high-concentrations of Yanghe Decoction respectively.DAPI staining and flow cytometry were used to detect cell cycle and apoptosis,qRT-PCR was used to detect cell signal transducer and activator of transcription(STAT)1,p53 mRNA expression,Western blot was used to detect STAT1/p53 signaling pathway related proteins STAT1,STAT3,p53 and Bax protein expressions.Results Compared with the blank control group,the apoptosis rate of Yanghe Decoction low-,medium-and highdosage groups significantly increased(P<0.01),the proportion of cells in G0/G1 phase increased,the proportion of cells in G2/M phase decreased(P<0.05,P<0.01),the expressions of STAT1 and p53 mRNA in Yanghe Decoction highdosage group significantly increased(P<0.01),the expressions of STAT1,p53 and Bax proteins in Yanghe Decoction medium-and high-dosage groups significantly increased(P<0.05,P<0.01),and the expression of STAT3 protein in Yanghe Decoction low-,medium-and high-dosage groups significantly decreased(P<0.01).Conclusion Yanghe Decoction can induce apoptosis of BT549 cells,and the mechanism is related to regulating the expression of STAT1/p53 signaling pathway related factors STAT1,STAT3,p53 and Bax.
