摘要
目的探讨葡甘露聚糖凝胶治疗混合感染性阴道炎大鼠的作用。方法将70只大鼠使用随机数表法分为空白组、模型组、葡甘露聚糖凝胶高、中、低剂量组(1.2、0.6、0.3 g/kg)、复方甲硝唑组(0.4 g/kg)、凝胶基质组,每组各10只。选用白色念珠菌、大肠杆菌、金黄色葡萄球菌建立大鼠混合感染性阴道炎模型,观察葡甘露聚糖凝胶对阴道炎的局部治疗作用,空白组不做处理。造模结束后开始局部给药治疗,治疗结束后,酶联免疫吸附测定法(ELISA法)检测大鼠血清中肿瘤坏死因子-α(TNF-α)、白介素(IL-1β、IL-6、IL-8)、表皮生长因子(EGFR)的含量;苏木精-伊红(HE)染色法观察阴道黏膜组织病理形态学变化。结果模型组大鼠血清中TNF-α、IL-1β、IL-6、IL-8大于空白组大鼠,差异有统计学意义(P<0.01),模型组血清中EGFR含量明显小于空白组大鼠,差异有统计学意义(P<0.01);给药后,葡甘露聚糖凝胶各剂量组、复方甲硝唑组血清中TNF-α、IL-1β、IL-6、IL-8均小于模型组大鼠,差异有统计学意义(P<0.01),EGFR含量均大于模型组大鼠,差异有统计学意义(P<0.01);阴道黏膜组织病理结果显示,模型组阴道黏膜固有层水肿,结缔组织排列疏松,伴较多炎性细胞散在浸润,与空白组比较,炎症反应明显加重,葡甘露聚糖凝胶高、中剂量组阴道被覆鳞状上皮,上皮细胞排列紧密,形态正常,固有层胶原丰富,排列紧密,见极少量炎性散在浸润。结论葡甘露聚糖凝胶对混合感染性阴道炎大鼠进行局部治疗后具有良好的效果。
Objective To investigate the therapeutic effect of glucomannan gel on mixed vaginitis in rats.Methods A total of 70 rats were divided into the blank group,the model group,the high,medium and low dose groups of glucomannan gel(1.2,0.6,0.3 g/kg),the compound metronidagole vaginal suppositories group(0.4 g/kg)and the gel matrix group using the random number table method,with 10 rats in each group.Candida albicans,Escherichia coli and Staphylococcus aureus were selected to establish a rat model of mixed vaginitis,and the local therapeutic effect of glucomannan gel on vaginitis was observed.The blank group was not treated.After the completion of modeling,local drug administration was started.After the completion of treatment,enzyme-linked immunosorbent assay(ELISA)was used to detect the tumor necrosis factor-α(TNF-α),interleukin(IL-1β,IL-6,IL-8)and epidermal growth factor receptor(EGFR)in rat serum;The hematoxylin and eosin(HE)staining was utilized to observe the histopathological changes of vaginal mucosa.Results The levels of TNF-α,IL-1β,IL-6 and IL-8 in the serum of rats in the model group were greater than those in the blank group,with statistically significant differences(P<0.01).The content of EGFR in the serum of the model group was significantly lower than that of the blank group,with statistically significant difference(P<0.01).After dose administration,the levels of TNF-α,IL-1β,IL-6 and IL-8 in the serum of rats in each dose group of glucomannan gel and the compound metronidagole vaginal suppositories group were lower than those in the serum of the model group,with statistically significant differences(P<0.01),while the content of EGFR was higher than that in the serum of rats in the model group,with statistically significant difference(P<0.01).The histopathological results of vaginal mucosa showed that the model group had edema in the lamina propria of vaginal mucosa,loose connective tissue arrangement,and scattered infiltration of more inflammatory cells.Compared with that seen in the blank group,th
