摘要
本研究拟对萝卜角果特异突变体开展研究,以萝卜长角果long silique(ls)突变体及萝卜野生型为研究材料,结合形态学及分子标记检测等技术探索上述萝卜角果突变体突变原因。经过多代自交纯化,基于EMS诱变获得的萝卜长角果突变体已表现稳定,实验分析表明,长角果突变体萝卜的角果平均长度为20.50 cm,正常角果萝卜角果平均长度为9.10 cm;长角果突变体萝卜的胚珠平均数量为9.5,正常角果萝卜的胚珠平均数量为4.5。通过PCR检测发现HZ001、SRC9-022、OI12F11等角果相关SSR分子标记及LS1和LS2基因表达在正常角果萝卜与长角果萝卜中表现出明显差异。上述研究结果表明,业已发现的萝卜长角果突变体表型特异且稳定,为萝卜种子产量改良提供优良研究材料。
In this study,we intended to carry out the research on the radish silique mutants including the radish ls mutants with longer silique and radish wild-type by comparing with morphological and molecular marker detection technology.The results showed that the mutant obtained by EMS treatment had stable characters as below:the length of the mutant silique was 20.50 cm,that of the normal one was 9.10 cm;the ovule number in one silique of the mutant was 9.5,and that of the normal one was 4.5.By PCR detection,we found that there were abundant differences between normal and long silique radish in HZ001,SRC9-022,OIL2F11 SSR molecular markers and gene expression of LSI and LS2.Those results showed that the mutant of radish with longer silique was specific and stable,which provided excellent research materials for the improvement of radish with high yield of seed.
