CADM-1对人宫颈癌Hela细胞转录组学的影响及意义

Effect and significance of CADM-1 on the transcriptome of human cervical cancer Hela cells

摘要 目的 探讨粘附分子CADM-1基因对人类宫颈癌细胞系Hela细胞转录组学的影响。方法 构建rLV-hCADM-1-ZsGreen-Puro慢病毒和rLV-ZsGreen-Puro对照慢病毒,将感染rLV-h CADM-1-ZsGreen-Puro慢病毒的Hela细胞作为rLV-CADM-1组,将感染rLV-ZsGreen-Puro对照慢病毒的Hela细胞作为rLV组。筛选差异基因集并进行基因转录能力、基因本体(GO)功能、京都基因与基因组数据库(KEGG)信号通路、Reactome通路及疾病本体(DO)功能分析。结果 rLV-CADM-1组CADM-1相对表达量(38.57±2.58)显著高于rLV组(0.90±1.08),差异有统计学意义(t=20.460,P<0.05)。基因差异分析共筛选出321个差异表达的基因,其中表达上调171个、表达下调150个。差异最显著的前10个表达上调基因包括OLFM1、TUSC3、ENSG00000285779、SPARC、CCL5、ENSG00000261512、CADM-1、OAS1、CLIC3及CSAG3。差异最显著的前10个表达下调基因包括COL16A1、SYTL4、ENSG00000278996、CGA、SLC47A1、TNC、ENSG00000276570、UGT3A1及MARCOL。功能分析结果显示,CADM-1改变了Hela细胞部分基因的转录能力、参与调控GO功能、KEGG信号通路、Reactome通路及DO功能。结论 CADM-1可明显影响Hela细胞的转录组学。 Objective To investigate the effect of CADM-1 gene on the transcriptome of human cervical cancer cell line Hela.Methods rLV-hCADM-1-ZsGreen Puro lentivirus and rLV-ZsGreen Puro control lentivirus were constructed. Hela cells infected with rLV-hCADM-1-ZsGreen Puro lentivirus were used as rLV CADM-1 group, and Hela cells infected with rLV-ZsGreen Puro control lentivirus were used as rLV group. The differential gene set was screened and gene transcription ability, gene ontology(GO) function, Kyoto Gene and Genome Database(KEGG) signal pathway, Reactome pathway and disease ontology(DO) function were analyzed.Results The relative expression of CADM-1 in rLV-CADM-1 group(38.57±2.58) was significantly higher than that in rLV group(0.90±1.08)(t=20.460, P<0.05). 321 differentially expressed genes were screened by gene difference analysis, including 171 up-regulated genes and 150 down-regulated genes. The top 10 up-regulated genes with the most significant differences include OLFM1, TUSC3, ENSG000000285779, SPARC, CCL5, ENSG00000261512, CADM-1, OAS1, CLIC3 and CSAG3. The first 10 down regulated genes with the most significant difference include COL16A1, SYTL4, ENSG000000278996, CGA, SLC47A1, TNC, ENSG000000276570, UGT3A1 and MARCOL. Functional analysis showed that CADM-1 changed the transcription ability of some genes in Hela cells, participated in regulating GO function, KEGG signal pathway, Reactome pathway and DO function.Conclusion CADM-1 can significantly affect the transcriptome of Hela cells.
