短链菊粉与高脂饲粮对不同基因型小鼠血清葡萄糖含量及盲肠微生物区系关联性的影响 被引量:1

Effects of Short Chain Inulin and High Fat Diet on Association of Serum Glucose Content and Cecal Microflora in Mice with Different Genotypes

摘要 本试验旨在研究短链菊粉与高脂饲粮对不同基因型小鼠血清葡萄糖含量及盲肠微生物区系关联性的影响。将8周龄40只雄性野生型C57BL/6J小鼠与40只雄性瘦素(Leptin)基因敲除小鼠分为8组,每组10只,单笼饲养。1~4组为野生型小鼠的不同处理组:野生型对照组(CT组)、野生型+10%短链菊粉组(CI组)、野生型+高脂组(CH组)、野生型+高脂+10%短链菊粉组(CHI组);5~8组为Leptin基因敲除小鼠的不同处理组:Leptin基因敲除对照组(OT组)、Leptin基因敲除+10%短链菊粉组(OI组)、Leptin基因敲除+高脂组(OH组)、Leptin基因敲除+高脂+10%短链菊粉组(OHI组)。试验期为8周,期间监测采食量与体增重。于第8周腹腔注射葡萄糖,注射后15、30、60和120 min测定血清葡萄糖含量。之后脱臼处死,测定体重与体长,取肝脏、生殖腺脂肪称重。取盲肠段及内容物测定微生物群落组成。结果表明:1)第12天CHI组体重显著低于CI组(P<0.05),第42~54天CH组体重显著低于CI组(P<0.05),Leptin基因敲除组间体重无显著差异(P>0.05)。野生型组体重均显著低于Leptin基因敲除组(P<0.05)。野生型组肥胖系数、肝重比基本低于Leptin基因敲除小组,OHI组的肝重比显著高于OI组和OT组(P<0.05)。2)30 min时,CH组和CHI组血清葡萄糖含量显著高于CT组和CI组(P<0.05);60 min时,OH组和OHI组血清葡萄糖含量分别显著高于OT组和OI组(P<0.05)。3)盲肠乳酸杆菌属(Lactobacillus)相对丰度与肝重比和Lee’s指数存在极显著正相关(P<0.01),而未命名梭菌纲_UCG-014属(norank_f_norank_o_Clostridia_UCG-014)和杜博西拉属(Dubosiella)相对丰度分别与肝重比和Lee’s指数存在极显著负相关(P<0.01),血清葡萄糖含量与布劳特氏菌属(Blautia)、拉克氏梭状芽孢杆菌属(Lachnoclostridium)、粪杆菌属(Faecalibaculum)和杜博西拉属相对丰度存在极显著正相关(P<0.01),而与毛螺菌科_NK4A136群(Lachnospiraceae_NK4A136_group)、未命名梭菌纲_ This experiment was conducted to investigate the effects of short chain inulin and high fat diet on the association of serum glucose content and cecal microflora in mice with different genotypes.Forty wild-type C57BL/6J male and forty Leptin gene knockout male mice at 8 weeks of age were divided into 8 groups,each group had 10 mice.Single cage breeding.Groups 1 to 4 were different treatment groups of wild-type mice:wild wild-type control group(CT group),wild-type+10%short chain inulin group(CI group),wild-type+high fat group(CH group),wild-type+high fat+10%short chain inulin group(CHI group);groups 5 to 8 were different treatment groups of Leptin gene knockout mice:Leptin gene knockout control group(OT group),Leptin gene knockout+10%short chain inulin group(OI group),Leptin gene knockout group+high fat group(OH group),Leptin gene knockout+high fat+10%short chain inulin group(OHI group).The trial lasted for 8 weeks,during which the feed intake and weight gain were monitored.At the 8th week,mice were intraperitoneally injected with glucose,and serum glucose content was measured before injection,15,30,60 and 120 min after injection.Then,the body weight and body length were measured.Liver and gonad fat were taken and weighed.The cecum segment and its contents were taken to determine the composition of microbial community.The results showed as follows:1)the body weight in CHI group was significantly lower than that in the CI group on day 12(P<0.05),the body weight in CH group was significantly lower than that in the CI group on days 42 to 54(P<0.05),there was no difference in body weight among the Leptin gene knockout groups(P>0.05).The body weight in the wild-type group was significantly lower than that in the Leptin gene knockout group(P<0.05).The obesity coefficient and liver to weight ratio in the wild-type group were lower than those in the Leptin gene knockout group,and the liver to weight ratio in OHI group was significantly higher than that in the OI and OT groups(P<0.05).2)The serum glucose content in CH and
