摘要 试验旨在研究甘草细胞总黄酮的提取方法及其抗氧化活性。以甘草细胞为试验材料,选取超声时间、液料比、乙醇浓度进行单因素及响应面试验,评价甘草细胞总黄酮粗提液的抗氧化性能。结果显示:甘草细胞总黄酮提取的最佳工艺条件为:超声时间90.70 min、液料比101.22 mL/g、乙醇浓度73.84%,此条件下,甘草细胞总黄酮含量为1.332%。抗氧化活性试验表明,总黄酮粗提液的总抗氧化能力优于维生素C(VC),对OH自由基的IC50值为136.78 mg/L,对DPPH自由基的IC_(50)值为44.87 mg/L,对ABTS自由基的IC_(50)值为2.01 mg/L。研究表明,甘草细胞总黄酮粗提液具有较好的抗氧化活性,试验可为药用植物活性成分在饲料领域的应用提供参考。 The study was to study the extraction method and antioxidant activity of total flavonoids from Glycyrrhiza uralensis Fisch.cells.Using glycyrrhiza cyrrhiza cells as experimental materials,single factor and response surface tests were conducted by ultrasonic time,liquid to solid ratio and ethanol concentration,and the antioxidant properties of total flavonoids crude extract from glycyrrhiza cyrrhiza cells were evaluated.The results showed that the optimal extraction conditions were ultrasonic time of 90.70 min,liquid to solid ratio of 101.22 mL/g,ethanol concentration of 73.84%.Under these conditions,the total flavonoids content of licorice cells was 1.332%.The antioxidant activity test showed that the total antioxidant capacity of total flavonoids extract was better than that of VC,and the IC50value of OH radical was 136.78 mg/L,DPPH radical IC_(50)value was 44.87 mg/L,and ABTS radical IC_(50)value was 2.01 mg/L.The study indicates that the crude extract of total flavonoids from Glycyrrhiza uralensis Fisch.cells has good antioxidant activity.The study can provide reference for the application of active ingredients from medicinal plants in the field of feed.
