摘要
本试验以白菜型春油菜品种——‘天祝小油菜’为试验材料,研究不同花蕾部位、不同诱导培养基及不同低温等预处理时间对花药愈伤组织诱导率的影响。无预处理时,筛选不同花蕾部位接种到不同诱导培养基的最佳组合,结果表明:以主花序蕾盘中盘青绿色、长度2~3 mm的花药为接种材料,在添加1.5 mg/L2,4-D、1 mg/L KT的B5培养基上培养,其花药愈伤组织诱导率最高为27.26%,均高于内盘、外盘花蕾花药与其他诱导培养基组合诱导率。预处理结果表明:4℃预处理4 d对花药愈伤组织诱导最有效,诱导率为60.27%,显著高于同期对照1.21倍;0.2 mg/L甘露醇培养4 d对花药愈伤组织诱导率为48.07%,显著高于对照和其他甘露醇溶液处理。花蕾在0.1 mol/L蔗糖溶液浸泡处理30 min后,花药愈伤组织诱导率为54.8%,显著高于对照和其他蔗糖溶液处理。综上以白菜型油菜主花序蕾盘、中盘青绿色,长度2~3 mm的花药为接种材料,4℃预处理4 d后接种在添加1.5 mg/L 2,4-D、1 mg/L KT的B5培养基上愈伤组织诱导效果最好。
In this experiment,to study the effects of different treatments including different bud parts,induction media,and low temperature treatment time on anther callus induction rate,the investigation was carried out using’Tianzhuxiaoyoucai’Brassica rapa L.as experimental material.The results showed that 4℃pretreatment for 4days was the most effective for anther callus inductionwiththe induction rateof 60.27%,which was 1.21 times higher than that of the control at the same period;0.2 mg/L mannitol for 4 days was 48.07%,which was significantly higher than that of other mannitol solutions and the control;the anther callus induction rate was 54.8%after the buds were soaked in 0.1 mg/L sucrose solution for 30 minutes.In conclusion,the best callus induction effect was obtained when the main inflorescence bud disc was green and the length of anther was 2~3 mm,and 1.5 mg/L2,4-D and 1 mg/L KT were added to B5 medium.The results of this study provide a theoretical reference for plant genetic transformation and haploid breeding of Brassica rapa L..
