麦冬提取物对心力衰竭大鼠心功能的影响 被引量:10

Effect of Ophiopogon japonicus extract on the cardiac function of rats with heart failure

摘要 目的探讨麦冬提取物调控环腺苷酸(cAMP)/腺苷一磷酸活化蛋白激酶(AMPK)信号通路对心力衰竭大鼠心功能的影响。方法将大鼠分为对照组、模型组、低剂量实验组(250 mg·kg^(-1)麦冬提取物)、高剂量实验组(500 mg·kg^(-1)麦冬提取物)及抑制剂组(500 mg·kg^(-1)麦冬提取物+20 mg·kg^(-1)Compound C),干预4周。以超声心动图检测左心室收缩末期内径(LVESD)、左心室舒张末期内径(LVEDD)、短轴收缩率(FS)、射血分数(EF)及心脏指数,以蛋白质印迹法检测心肌组织中蛋白激酶A(PKA)、p-PKA、cAMP反应元件结合蛋白(CREB)、p-CREB、AMPK、p-AMPK蛋白表达。结果对照组、模型组、抑制剂组和低、高剂量实验组LVESD分别为(3.60±0.18),(6.31±0.27),(5.84±0.24),(5.26±0.22),(4.39±0.24)mm,LVEDD分别为(5.58±0.22),(7.72±0.32),(7.16±0.21),(6.70±0.28),(6.13±0.26)mm,心脏指数分别为(2.81±0.12),(4.13±0.17),(3.88±0.13),(3.69±0.18),(3.25±0.15)g·kg^(-1),FS分别为(20.02±0.45)%,(17.85±0.23)%,(18.30±0.25)%,(18.74±0.28)%,(19.45±0.32)%,EF分别为(64.86±3.52)%,(29.34±2.08)%,(35.54±2.85)%,(40.67±3.04)%,(50.18±2.96)%,模型组与对照组比较,低、高剂量实验组与模型组比较,抑制剂组与高剂量实验组比较,差异均有统计学意义(均P<0.05)。结论麦冬提取物可能通过激活cAMP/AMPK信号通路,改善心力衰竭大鼠心功能。 Objective To investigate the effect of Ophiopogon japonicus extract on cardiac function in rats with heart failure by regulating the cyclic adenosine monophosphate(cAMP)/adenosine monophosphate-activated protein kinase(AMPK)signaling pathway.Methods Rats were devided into control group,model group,low dose experimental group(250 mg·kg^(-1)Ophiopogon japonicus extract),high dose experimental group(500 mg·kg^(-1)Ophiopogon japonicus extract)and inhibitor group(500 mg·kg^(-1)Ophiopogon japonicus extract+20 mg·kg^(-1)Compound C),intervention for 4 weeks.Echocardiography was used to detect the left ventricular end systolic diameter(LVESD),left ventricular end diastolic diameter(LVEDD),fraction shortening(FS),ejection fraction(EF)and cardiac index.Western Blot was used to detect protein kinase A(PKA),p-PKA,cAMP response element binding protein(CREB),p-CREB,AMPK,p-AMPK protein expression in myocardial tissue.Results LVESD in control group model group,inhibitor group and low dose,high dose experimental groups were(3.60±0.18),(6.31±0.27),(5.84±0.24),(5.26±0.22),(4.39±0.24)mm;LVEDD were(5.58±0.22),(7.72±0.32),(7.16±0.21),(6.70±0.28),(6.13±0.26)mm;heart index were(2.81±0.12),(4.13±0.17),(3.88±0.13),(3.69±0.18),(3.25±0.15)g·kg^(-1);FS were(20.02±0.45)%,(17.85±0.23)%,(18.30±0.25)%,(18.74±0.28)%,(19.45±0.32)%;EF were(64.86±3.52)%,(29.34±2.08)%,(35.54±2.85)%,(40.67±3.04)%,(50.18±2.96)%,there were statistically significant differences between model group and control group,between low,high dose experimental group and model group,between inhibitor group and high dose experimental group(all P<0.05).Conclusion Ophiopogon japonicus extract may activate the cAMP/AMPK signaling pathway to improve the cardiac function of rats with heart failure.
