基于UPLC指纹图谱结合化学计量学的肝爽颗粒质量评价研究 被引量:1

摘要 目的:建立肝爽颗粒的UPLC指纹图谱,并结合化学计量学等方法寻找表征不同批次肝爽颗粒质量差异性标志物,为科学全面地评价肝爽颗粒的质量提供参考。方法:采用UPLC法对18批肝爽颗粒进行分析,建立肝爽颗粒指纹图谱,使用“中药色谱指纹图谱相似度评价系统”进行相似度评价,利用对照品对共有峰进行指认,并结合化学计量学方法对不同批次肝爽颗粒质量及控制方法进行分析和评价。结果:建立的指纹图谱确定23个共有峰,指认出10个成分,分别为阿魏酸(5号峰)、虎杖苷(6号峰)、菊苣酸(7号峰)、柚皮苷(11号峰)、橙皮苷(12号峰)、迷迭香酸(13号峰)、新橙皮苷(14号峰)、丹酚酸B(16号峰)、大黄素(21号峰)、大黄素甲醚(23号峰);18批肝爽颗粒指纹图谱的相似度均在0.990以上;聚类分析和主成分分析(PCA)将18批肝爽颗粒基本分为3类;正交偏最小二乘法-判别分析(OPLS-DA)筛选得到12个变量重要性投影(VIP)>1的共有峰(峰7、2、19、22、3、20、14、5、1、11、13、4),经对照品指认了其中5个差异性标志物(菊苣酸、新橙皮苷、阿魏酸、柚皮苷、迷迭香酸)。结论:该研究建立的UPLC指纹图谱方法合理、有效、准确、简便,结合化学计量学分析方法可用于评价肝爽颗粒的质量,为肝爽颗粒的质量控制提供科学依据。 Objective: To establish the UPLC fingerprint of Ganshuang granules and determine the markers used to characterize the quality difference of Ganshuang granules with different batches coupled with chemometrices, so as to provide reference for scientific and comprehensive evaluation of the quality of Ganshuang granules. Methods: A total of 18 batches of samples were analyzed to establish the UPLC fingerprint of Ganshuang granules. Similarity evaluation was performed by using Traditional Chinese Medicine Chromatographic Fingerprint Similarity Evaluation System to confirm the common peaks, which were identified by comparing with reference substances. Furthermore, chemometrics was used to analyze and evaluate the quality of Ganshuang granules with different batches. Results: The UPLC fingerprint was established and 23 common peaks were identified in the study. And ten common peaks among them were identified as ferulic acid(peak 5), polydatin(peak 6), chicoric acid(peak 7), naringin(peak 11), hesperidin(peak 12), rosmarinic acid(peak 13), neohesperidin(peak 14), salvianolic acid B(peak 16), emodin(peak 21), and emodin-3-methyl ether(peak 23);the similarity of fingerprints of 18 batches of Ganshuang granules was above 0.990;cluster analysis and principal component analysis(PCA) divided the 18 batches of samples into 3 categories;discriminant analysis(OPLS-DA) of the 18 batches of Ganshuang granules screened 12 common peaks with VIP>1(peaks 7, 2, 19, 22, 3, 20, 14, 5, 1, 11, 13, 4), and five of them(chicoric acid, neohesperidin, ferulic acid, naringin, rosmarinic acid) could be served as differential markers.Conclusion: The established UPLC fingerprint method is reasonable, efficient, accurate and simple combined with chemometrics, which can be used to evaluate the quality of Ganshuang granules and provide scientific basis for the quality control of Ganshuang granules.
