多效唑对番茄上胚轴矮化的影响 被引量:7

Effects of paclobutrazole on epicotyl dwarfing in tomato

摘要 夏秋季节,园艺设施内易出现高温、昼夜温差小等现象,致使番茄幼苗出现徒长的现象。缩短上胚轴可以抑制徒长,提高产量,降低生产成本。多效唑是内源赤霉素(GA)合成的抑制剂,能够延缓作物生长。本文利用不同浓度多效唑喷施长节间番茄幼苗品系DH,研究多效唑矮化番茄上胚轴的作用机理。结果发现,200 mg·L^(-1)多效唑处理后的上胚轴生长率和株高显著降低,叶绿素a、b含量、单果重、果实中可溶性蛋白、抗坏血酸和可溶性固形物含量均显著升高。多效唑处理5 d后,与未处理的植株相比,内源GA和生长素(IAA)含量显著增加了58.41%和16.08%,GA、IAA代谢通路相关基因的表达量也显著提高。根据上胚轴内部细胞形态观察发现,与未处理的植株相比,多效唑处理后的上胚轴内细胞数量增加,且细胞体积变小。说明喷施多效唑后,番茄上胚轴受到外源物的刺激,致使机体内源IAA和GA含量增加,以维持细胞激素水平的稳态,最终导致细胞数量增加。本文结果为设施番茄苗期徒长的控制和幼苗质量的提高、化控技术的应用提供参考,同时为促进番茄理想株型性状的改良提供重要的实践意义。 In the seasons of summer and autumn,horticultural facilities are prone to have high temperature and small temperature difference between day and night,resulting in serious leggy of tomato(Solanum lycopersicum)seedlings.Shortening the epicotyl of tomato could restrain the elongation,increase the yield,and reduce the production cost.Paclobutrazole is an inhibitor of endogenous gibberellin(GA)synthesis and can delay crop growth.In this experiment,different concentrations of paclobutrazol were spaying on long internode tomato seedling strain DH to study the mechanism of paclobutrazol dwarfing tomato epicotyl.The results showed that the growth rate of epicotyl and plant height were significantly reduced,the contents of chlorophyll a and b weight per fruit,soluble protein,ascorbic acid and soluble solid of fruits were significantly increased by 200 mg·L^(-1)paclobutrazole treatment.After 5 d of paclobutrazole treatment,compared with the without paclobutrazole treatment,the content of endogenous GA and auxin(IAA)were increased by 58.41%and 16.08%,respectively,and the expression levels of genes related to GA and IAA metabolic pathway also increased significantly.Based on cell morphology in epicotyl,the number of the cell were increased and the size of cell were decreased.The results showed that spraying paclobutrazole on tomato epicotyl resulted in the increase of GA and IAA contents due to the stimulation of exogenous substances,to maintain the homeostasis of hormone levels in the cell,leading to an increase in the number of cells increase.The results of this study will provide references for the control of seedling leggy,the improvement of seedling quality and the application of chemical control technology of greenhouse tomato.Meanwhile,it provides important practical significance for improving tomato ideal plant type traits.
