基于Web服务的禽流感病毒基因分析软件FluSoft的创建与应用

摘要 解读病毒基因组信息是探明病毒关键分子特性的重要步骤。然而,解码基因信息,需要在较强的生物信息学背景下,查阅大量文献、选择参考序列、开展序列比对、进行遗传进化分析、验证并集成数据信息等,其过程工作量大、耗时、技术含量高。为解决上述问题,采用Perl语言集成一系列软件和数据集,创建了禽流感病毒基因分析软件(FluSoft),实现了禽流感病毒基因组快速批量注释和分析。本软件通过Web网页对话框输入FASTA格式的禽流感病毒基因序列,可进行关键生物特性和遗传进化分析,实现了以下功能:(1)注释查询序列的基因片段/亚型(针对H5或H7亚型,增加展示其HA基因裂解位点氨基酸序列及碱性氨基酸个数)及基因突变的生物学意义,如耐药性、宿主受体特异性、毒力、糖基化位点及在家禽或哺乳动物中的传播能力等;(2)注释查询序列在命名系统中最密切相关的演化分支。综上所述,FluSoft软件(http://flusoft.hqhuitong.com)为禽流感流行病学调查监测、流行特征分析及风险预警提供了有力的技术支撑。 Decoding information hiding in the viral genome was an important step to identify the key molecular characteristics of viruses.However,the procedure was very sophisticated because of heavy workload,time-consuming and complicated technology,as it required to refer to a large amount of literature,select reference sequences,conduct sequence alignment,analyze genetic evolution,verify and integrate data information under a strong bioinformatics background.In order to address the above problems,a series of software and data sets were integrated by Perl language,and an analysis software(FluSoft)for avian influenza virus(AIV)genomes was established,which could be used for rapid batch annotation and analysis of AIV genomes.The software could be used to analyze key biological characteristics and genetic evolution through inputting the gene sequence of AIV in FASTA format into the dialog box on the Web,by which the following functions were performed:(1)the biological significance of gene mutations and query sequences'gene segment/subtype(for H5 and H7 subtypes,the sequence of the HA cleavage site and its number of basic amino acids were increased)were annotated,such as drug resistance,specificity of host receptors,virulence,glycosylation sites and their transmission capacity in poultry or mammals;(2)the most closely related evolutionary branch in the nomenclature system was annotated.In conclusion,future epidemiological investigation and monitoring as well as analysis on the prevalence characteristics and risk early warning of avian influenza were technically supported by the FluSoft software.
