摘要
试验旨在研究牛乳外泌体对葡聚糖硫酸钠(DSS)诱导的结肠炎小鼠的影响。选取21日龄SPF级雄性BALB/c小鼠42只,随机分为3组,分别为对照组、模型组(DSS组)、牛乳外泌体组(Exo组),每组7个重复,每个重复2只。试验期间对照组小鼠自由饮水;DSS组小鼠前5 d自由饮用3.5%的DSS溶液,后7 d自由饮水,Exo组小鼠前5 d自由饮用3.5%的DSS溶液,后7 d每只小鼠灌喂10μL牛乳外泌体(1 g/L)。试验期12 d。结果显示:各组小鼠的平均日增重均差异不显著(P>0.05),DSS组和Exo组小鼠的平均日采食量显著低于对照组(P<0.05)。DSS组和Exo组小鼠血清白细胞介素-6(IL-6)含量显著升高(P<0.05),白细胞介素-4(IL-4)、白细胞介素-10(IL-10)和肿瘤坏死因子-α(TNF-α)含量均差异不显著(P>0.05)。各组小鼠血清的抗氧化指标和肠道长度均差异不显著(P>0.05)。试验第12 d,Exo组小鼠的DAI评分显著低于DSS组(P<0.05)。研究表明,灌喂10μL牛乳外泌体(1 g/L)对DSS诱导结肠炎的小鼠影响有限,可能是由于灌喂的剂量较低。
The experiment was to investigate the effect of milk exosomes on colitis mice induced by sodium dextran sulfate(DSS). Forty-two 21-day-old SPF male BALB/c mice were randomly divided into three groups: Control group,model group(DSS group) and milk exosome group(Exo group), with seven replicates in each group and two mice in each replicate. The mice in control group drank water freely during the experiment. Mice in DSS group were given 3.5% DSS solution for the first 5 d, and drank water for the last 7 d. Mice in Exo group were given 3.5% DSS solution for the first5 d, and each mouse was fed with 10 μL milk exosomes(1 g/L) for the last 7 d. The experiment lasted for 12 d. The results showed that there was no significant difference in average daily gain among all groups(P>0.05), and the average daily feed intake of mice in DSS group and Exo group was significantly lower than that in control group(P<0.05). Serum interleukin-6(IL-6) content was significantly increased in DSS group and Exo group(P<0.05), but there was no significant difference in serum interleukin-4(IL-4), interleukin-10(IL-10) and tumor necrosis factor-α(TNF-α)(P>0.05). There was no significant difference in serum antioxidant indexes and intestinal length among all groups(P>0.05). On 12 d, DAI score of Exo group was significantly lower than that of DSS group(P<0.05). The study indicates that 10 μL bovine milk exosomes(1 g/L) has limited effect on DSS induced colitis mice, possibly due to the low dose.
