药用植物黄芩苷对COPD鼠的炎症和氧化应激的影响 被引量:2

Effects of Baicalin on Inflammation and Oxidative Stress in COPD Rat

摘要 黄芩(Scutellaria baicalensis Georgi)是一种传统的中药,从黄芩的干燥根中提取的黄酮类化合物黄芩苷具有多种药用价值,其中包括抗炎、抗菌、解热、镇痛、降压等。已有报道认为黄芩苷在香烟烟雾诱导的哮喘小鼠气道炎症和气道重塑中具有抗炎作用,但黄芩苷是否通过SIRT1/PGC-1α信号途径减轻COPD肺炎症和氧化应激尚不明晰。本研究试图揭示黄芩苷对慢性阻塞性肺疾病(COPD)老年大鼠炎症和氧化应激的影响,以及黄芩苷对Sirtuin1(SIRT1)和过氧化物增殖物活化受体γ辅激活因子-1α(PGC-1α)的激活作用。研究结果显示,与COPD组相比,黄芩苷组的细胞浸润程度明显减轻、炎症细胞数量减少、肺泡囊和肺泡间隙减小。黄芩苷组的总细胞数和中性粒细胞百分比显著减少(P<0.05)。而且,黄芩苷组的血清IL-6和IL-8的水平明显低于COPD组(P<0.05);与COPD组相比,黄芩苷组的MDA水平显著降低而SOD活性显著增加。同时,黄芩苷组的SIRT1和PGC-1αmRNA和蛋白表达增加(P<0.05)。本研究结果初步表明黄芩苷可有效抑制COPD老年大鼠的肺部炎症反应和氧化应激。黄芩苷通过激活SIRT1/PGC-1α信号途径来抑制炎症和氧化应激。 Huangqin(Scutellaria baicalensis Georgi)is a traditional Chinese medicine.The flavonoid baicalin extracted from the dried root of Huangqin has many medicinal values,including anti-inflammatory,antibacterial,antipyretic,analgesic,hypotensive,etc.It has been reported that baicalin has anti-inflammatory effect on airway inflammation and airway remodeling in asthmatic mice induced by cigarette smoke,but it is unclear whether baicalin passes SIRT1/PGC-1αsignal pathway alleviates inflammation and oxidative stress in COPD lung.This study attempts to reveal the effects of baicalin on inflammation and oxidative stress in aged rats with chronic obstructive pulmonary disease(COPD)and activation of baicalin on Sirtuin1(SIRT1)and peroxides proliferator activated receptorsγcoactivator factor-1α(PGC-1α).The results showed that compared with COPD group,baicalin group significantly reduced the degree of cell infiltration,the number of inflammatory cells,alveolar sacs and alveolar spaces.The total number of cells and the percentage of neutrophils in baicalin group decreased significantly(P<0.05).Moreover,the levels of serum IL-6 and IL-8 in baicalin group were significantly lower than those in COPD group(P<0.05).Compared with COPD group,the level of MDA in baicalin group was significantly decreased while the activity of SOD was significantly increased.Meanwhile,SIRT1 and PGC-1αof baicalin group mRNA and protein expression increased(P<0.05).The results of this study preliminarily showed that baicalin could effectively inhibit the pulmonary inflammatory response and oxidative stress of COPD aged rats.Baicalin activates SIRT1/PGC-1αsignal pathways to inhibit inflammation and oxidative stress.
