黄连解毒汤对脑梗死合并肠损伤大鼠SCF/C-kit信号通路的影响 被引量:5

Effect of Huanglian Jiedu Decoction on SCF/C-kit Signaling Pathway in Rats with Cerebral Infarction Complicated with Intestinal Injury

摘要 目的观察黄连解毒汤对脑梗死合并肠损伤大鼠干细胞因子(SCF)/酪氨酸激酶受体(C-kit)信号通路的影响。方法将80只健康Wistar10周龄雄性大鼠随机分为模型组、黄连解毒汤组、莫沙必利组、假手术组,并设有正常组。采用线栓法进行大脑中动脉梗死建模。黄连解毒汤组给予黄连解毒汤(27 mg/mL)混悬液,莫沙必利组给予莫沙必利(0.09 mg/mL)悬浊液,其余组给予1 m L生理盐水,连续给药7 d。各组分别在4、7 d时,采用HE染色法观察大鼠肠组织病理形态学变化,并进行病理损伤评分;采用实时荧光定量检测大鼠肠组织中SCF、C-kit m RNA表达水平;采用Western blotting检测大鼠肠组织中SCF、C-kit蛋白表达水平。结果与正常组、假手术组比较,模型组、黄连解毒汤组、莫沙必利组大鼠肠黏膜病理评分均明显升高(均P<0.01),肠组织SCF和C-kit mRNA、蛋白表达水平均明显降低(P<0.01);而正常组、假手术组间差异无统计学意义(P>0.05)。与模型组比较,黄连解毒汤组、莫沙必利组大鼠肠黏膜病理评分均显著降低(均P<0.01),肠组织SCF和C-kit mRNA、蛋白表达水平均明显升高(P<0.01),且与莫沙必利组比较,黄连解毒汤组大鼠肠黏膜病理评分均明显降低(P<0.05),肠组织SCF和C-kit mRNA、蛋白表达水平均明显升高(P<0.05)。结论脑梗死急性期易合并肠黏膜损伤。黄连解毒汤对脑梗死合并肠损伤大鼠的肠黏膜具有一定的保护作用。其机制可能与黄连解毒汤可升高大鼠肠组织中SCF、C-kit m RNA及蛋白的表达水平有关。 Objective:To investigate the effect of Huanglian Jiedu Decoction on SCF/C-kit signaling pathway in rats with cerebral infarction complicated with intestinal injury. Methods:A total of 80 healthy Wistar 10-weekold male rats were randomly divided into the model group,Huanglian Jiedu Decoction group,mosapride group,sham operation group,and normal group;the above groups were further divided into the 4 d group and 7 d group,and there were a total of 10 groups. Middle cerebral artery infarction model was established by suture method. The Huanglian Jiedu Decoction group was given Huanglian Jiedu Decoction(27 mg/mL) suspension,the mosapride group was given mosapride(0.09 mg/mL)suspension,and the mosapride group was given 1 m L saline. Each 4 d group was treated for 4 days,and the 7 d group was treated for 7 days. HE staining was used to observe the pathological morphological changes of intestinal tissue in rats,and the pathological damage score was performed. The expression levels of SCF and C-kit m RNA in rat intestinal tissues were detected by real-time fluorescence quantitative analysis. Western blotting was used to detect the expression levels of SCF and C-kit proteins in intestinal tissues of rats. Results:Compared with the normal group and the sham operation group,the pathological scores of intestinal mucosa of rats in the model group,Huanglian Jiedu Decoction group and mosapride group were significantly increased(P < 0.05). Compared with the model group,the pathological scores of intestinal mucosa in Huanglian Jiedu Decoction group and mosapride group were significantly decreased(P < 0.01),and the mRNA and protein expressions of SCF and C-kit in intestinal tissue were significantly increased(P < 0.01). Compared with mosapride group,the pathological scores of intestinal mucosa in Huanglian Jiedu Decoction group were significantly decreased(P < 0.05),and the mRNA and protein expressions of SCF and C-kit in intestinal tissue were significantly increased(P < 0.05).
