绵羊基因组DNA的提取及TLR基因片段的PCR扩增

摘要 Toll样受体(Toll-like receptors,TLRs)作为天然免疫系统中的重要分子,最突出的生物学功能是引发炎症反应,促进抗原递呈细胞的成熟,以此诱导机体获得性免疫反应。为研究TLR在疾病抗感染过程中起重要作用,提取绵羊基因组DNA,对TLR2和TLR4基因相关片段进行PCR扩增,得到品质较好的DNA和目的片段,并对试验过程中的影响因素进行讨论和总结,为进一步探索绵羊TLR2和TLR4基因片段等位基因的多态性及其与绵羊乳房炎之间的相关性奠定物质基础。 Toll-like receptors(TLRs),as an important molecule in the innate immune system,play an important biological role in triggering inflammatory response and promoting the maturation of antigen-presenting cells to induce the acquired immune response.Studies have shown that TLR play an important role in the process of disease resistance to infection.We extracted sheep genomic DNA,amplified TLR2 and TLR4 gene related fragments by PCR,got good quality DNA fragments and puro-se,and do to test the influence factors in the process of discussion and summary,in order to further explore the sheep TLR2 and TLR4 gene alleles polymorphism and its the correlation between sheep mastitis and lay the material foundation.
