新风胶囊通过调节Notch1通路、巨噬细胞极化改善类风湿关节炎大鼠关节炎症 被引量:3

Xinfeng Capsules Improving Joint Inflammation in Rats with Rheumatid Arthritis by Regulating Notch1 Pathway and Macrophage Polarization

摘要 目的:观察新风胶囊对类风湿关节炎大鼠关节炎症的影响,并初步探讨其作用机制。方法:将32只清洁级雄性SD大鼠随机分为正常组、模型组、甲氨蝶呤组(0.98 mg·L^(-1))和新风胶囊组(0.34 g·L^(-1)),每组8只,除正常组外,其余组大鼠右后足跖皮内注射弗氏完全佐剂构建类风湿关节炎大鼠模型。致炎后第12天开始灌胃给予相应药物,正常组及模型组给予10 mL·kg^(-1)生理盐水灌胃,给药30天后,检测各组大鼠足趾肿胀度和关节炎指数;HE染色观察大鼠关节滑膜组织病理形态变化;ELISA检测大鼠血清细胞因子白细胞介素-1(interleukin-1,IL-1)、IL-10、IL-23、转化生长因子-β(transforming growth factor-β,TGF-β)的表达;流式细胞术检测大鼠外周血炎症极化标志物CD80^(+)细胞、CD163^(+)细胞的表达;Western Blot检测大鼠关节滑膜组织中Notch1、Hes1蛋白的表达。结果:与正常组比较,模型组大鼠足趾肿胀度、关节炎指数明显升高(P<0.01),CD80^(+)细胞表达明显升高(P<0.01),CD163^(+)细胞表达明显降低(P<0.01),IL-1、IL-23、TGF-β表达水平明显升高(P<0.05或P<0.01),IL-10表达水平明显降低(P<0.01),滑膜组织中Notch1、Hes1蛋白表达水平明显升高(P<0.01),关节滑膜出现明显的病理变化;与模型组比较,新风胶囊组大鼠足趾肿胀度、关节炎指数明显降低(P<0.01),CD80^(+)细胞表达明显降低(P<0.01),CD163^(+)细胞表达明显升高(P<0.05),IL-1、IL-23、TGF-β表达水平明显降低(P<0.05),IL-10表达水平明显升高(P<0.01),滑膜组织中Notch1、Hes1蛋白表达水平显著降低(P<0.05),关节滑膜组织病理变化得到明显改善。结论:新风胶囊可能通过调节Notch1通路、巨噬细胞极化改善类风湿关节炎大鼠关节炎症。 Objective:To observe the effect and mechanism of Xinfeng capsule on joint inflammation in rats with Rheumatid arthritis.Methods:Thirty-two clean-grade male SD rats were randomly divided into normal group,model group,and methotrexate group(0.98 mg·L^(-1))and Xinfeng capsule group(0.34 g·L^(-1)),8 rats in each group,except for the normal group,the rats in the remaining groups were injected with Freund′s complete adjuvant in the right hindplantar plantar to construct a rat model of rheumatoid arthritis.On the 12th day after inflammation,the corresponding drugs were given by gavage,10 mL·kg^(-1) normal saline was given to the normal group and the model group,and the toe swelling and arthritis index of rats in each group were detected 30 days after administration.HE staining detected the histopathological morphological changes of the synovial membrane of rats.ELISA detected the expression of serum cytokines interleukin-1(IL-1),IL-10,IL-23,transforming growth factorβ(TGF-β)in rats.Flow cytometry detected the expression of CD80^(+)cells and CD163^(+)cells of peripheral blood inflammatory polarization markers in rat.Western Blot detected the expression of Notch1 and Hes1 proteins in the synovial tissue of rat joints.Results:Compared with the normal group,the toe swelling and arthritis index of rats in the model group were significantly increased(P<0.01),CD80^(+)cell expression was significantly increased(P<0.01),CD163^(+)cell expression was significantly decreased(P<0.01),IL-1,IL-23 and TGF-βexpression levels were significantly increased(P<0.05 or P<0.01),IL-10 expression levels were significantly reduced(P<0.01),and Notch1 and Hes1 protein expression levels in synovial tissues were significantly increased(P<0.01),pronounced pathological changes in the synovial membrane of the joint;Compared with the model group,the toe swelling and arthritis index of the Xinfeng capsule group were significantly reduced(P<0.01),the expression of CD80^(+)cells was significantly reduced(P<0.01),the expression of CD163^(+)cells was
