5-甲基胞嘧啶 5-羟甲基胞嘧啶在女性甲状腺乳头状癌组织中的表达及与TET蛋白的关系 被引量:1

Expression of 5-mC and 5-hmC in female papillary thyroid carcinoma and their relationship with TET protein

摘要 目的 探讨5-甲基胞嘧啶(5-mC)、5-羟甲基胞嘧啶(5-hmC)在女性甲状腺乳头状癌(PTC)组织中的表达及与TET蛋白的关系。方法 随机选取2019年1月—2020年1月于宁波大学附属人民医院进行手术治疗的33例女性PTC患者作为研究对象,经病理科收集甲状腺癌组织和癌旁组织(距离癌组织2~3 cm)。提取组织DNA,计算全基因组DNA甲基化率,比较PTC癌组织和癌旁组织中5-mC和5-hmC表达水平。提取组织RNA,反转录后利用荧光定量聚合酶链式反应,以GAPDH为参照,采用2-△△Ct表示组织中TET1、TET2、TET3蛋白表达水平,比较PTC癌组织与癌旁组织TET蛋白表达水平。结果 PTC癌组织5-mC和5-hmC表达水平低于癌旁组织,差异均有统计学意义(t=13.841、10.900,均P<0.05);PTC癌组织TET1、TET2和TET3蛋白mRNA表达水平低于癌旁组织,差异均有统计学意义(t=4.780、10.766、19.119,均P<0.05);PTC癌组织5-hmC表达水平与TET1蛋白呈正相关,差异有统计学意义(r=0.789,P<0.05)。结论 PTC癌组织中5-mC、5-hmC、TET1、TET2及TET3蛋白呈低表达,其中5-hmC表达水平与TET1蛋白呈正相关,5-hmC与TET1有可能成为PTC的诊断和治疗新靶点。 Objective To investigate the expression of 5-methylcytosine(5-mC) and 5-hydroxymethylcytosine(5-hmC) in female papillary thyroid carcinoma(PTC) and their relationship with TET protein.Methods 33 female PTC patients underwent surgery in our hospital from January 2019 to January 2020 were enrolled.The cancer tissues and adjacent normal tissues were collected in the pathology department.Global DNA methylation in tissue was determined.Expression levels of 5-mC and 5-hmC in cancer tissues and adjacent normal tissues were compared.Tissue RNA was extracted, and fluorescence quantitative polymerase chain reaction was used after reverse transcription.With GAPDH as the reference, 2-△△Ctwas used to indicate the expression levels of TET1, TET2 and TET3 proteins in tissues.Results The expression levels of 5-mC and 5-hmCin cancer tissues were lower than that in adjacent normal tissues(t=13.841、10.900, P<0.05).The mRNA expression levels of TET1, TET2 and TET3 proteins in cancer tissues were lower than that in adjacent normal tissues(t=4.780、10.766、19.119, P<0.05).The expression of 5-hmC in cancer tissues was positively correlated with TET1 protein(r=0.789, P<0.05).Conclusion In PTC tissues, the expression of 5-mC, 5-hmC, TET1, TET2 and TET3 are down-regulated, moreover, the expression of 5-hmC is positively correlated with TET1 protein.5-hmC and TET1 may become new targets for diagnosis and treatment of PTC point.
