摘要
目的运用网络药理学方法探讨鸡血藤治疗银屑病的作用机制。方法利用中药系统药理学数据库和分析平台筛选出鸡血藤的活性成分及作用靶点,利用GeneCards、OMIM数据库预测银屑病相关靶点。对药物靶点与疾病靶点取交集后获得共同靶点,将共同靶点及其对应的活性成分输入CytoScape 3.7.0软件,构建“药物-活性成分-靶点-疾病”可视化网络。针对共同靶点,利用STRING数据库构建蛋白-蛋白相互作用(PPI)网络图,并进行基因本体论功能富集分析和京都基因与基因组百科全书通路分析。结果筛选出24个鸡血藤活性成分,共获得共同靶点86个。药物-活性成分-靶点-疾病可视化网络中,度值排前4位的鸡血藤活性成分依次为木犀草素、芒柄花黄素、甘草查尔酮A、β谷甾醇。PPI网络中,度值排前5位的靶点依次为RAC-α丝氨酸/苏氨酸蛋白激酶、信号传导子及转录激活因子3、肿瘤抗原p53、转录因子AP-1、Caspase-3;鸡血藤治疗银屑病的主要潜在靶点参与了1442个生物学过程、67个细胞成分和115个分子功能,涉及脂质与动脉粥样硬化信号通路、PI3K/AKT信号通路、IL-17信号通路、乙型肝炎信号通路等184条信号转导通路。结论鸡血藤中的多种有效成分可通过多个作用靶点和信号通路发挥治疗银屑病的作用。
Objective To explore the mechanism of Spatholobus suberectus dunn in the treatment of psoriasis by the application of network pharmacology.Methods The active components and effect targets of Spatholobus suberectus dunn were screened by Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform.The targets related to psoriasis were predicted by the GeneCards and OMIM databases.After intersection between drug targets and diseases targets,the common targets were obtained,and the common targets and their corresponding active components were input into the CytoScape 3.7.0 software to construct the“drug-active component-target-disease”visual network.With respect to common targets,the protein to protein interaction(PPI)network was constructed,and the Gene Ontology function enrichment analysis and Kyoto Encyclopedia of Genes and Genomes pathway analysis were performed by the STRING database.Results A total of 24 active components of Spatholobus suberectus dunn were screened,and 86 common targets were obtained.In the visual network of drug-active components-targets-disease,the top 4 active components of Spatholobus suberectus dunn in degree value successively were luteolin,formononetin,licochalcone A and beta-sitosterol.In the PPI network,the top 5 targets in degree value successively were RAC-alpha serine/threonine protein kinase,signal transducer and activator of transcription 3,tumor antigen p53,transcription factor AP-1 and Caspase-3.The major potential targets of Spatholobus suberectus dunn for the treatment of psoriasis were involved in 1442 biological processes,67 cell components and 115 molecular functions,concerning 184 signaling transduction pathways as for lipid and atherosclerosis signaling pathway,PI3K/AKT signaling pathway,IL-17 signaling pathway and hepatitis B signaling pathway,etc.Conclusion A variety of effective components in Spatholobus suberectus dunn can play a role in the treatment of psoriasis through multiple effect targets and signaling pathways.
