摘要
青稞对青藏高原极端环境有极强的适应性。镉诱导的植物衰老会降低作物产量和质量,并严重威胁人类健康。本研究旨在探究镉胁迫下两种青稞叶片的衰老差异。将‘昆仑12’(晚衰型,late-stage senescing,LS)及‘肚里黄’(早衰型,early-stage senescing,ES)两种青稞进行液体培养后,用浓度为25、50、75、100、150μmol/L的氯化镉对其幼苗分别处理2 d,观察其生长情况,测定生理指标,比较其镉耐受性和衰老进程的差异。结果显示镉处理使二者表现出依赖Cd浓度增加的叶片发黄、叶尖卷曲、叶绿素含量降低、膜损伤增加、过氧化氢积累和镉积累等现象。镉处理引起两种品种中CAT活性的增加,不同的是在ES中CAT活性随镉浓度的增加先增加后降低,而在LS中呈持续增加的趋势,其余三种抗氧化酶SOD、POD和APX的活性变化为随Cd浓度的升高呈先升后降的趋势,且在ES中均显著高于LS。低浓度的Cd对LS的影响较小,而高浓度的Cd使LS和ES均表现出衰老,其衰老进程存在差异。镉浓度达75μmol/L时,对两种青稞的影响差异显著。说明镉诱导下,LS较ES表现出较强的镉耐受性,ES衰老进程明显被加快。本研究结果有助于后期深入研究镉诱导的青稞叶片衰老的具体机制。
Highland barley,showed strong adaptability in a variety of harsh environments in Qinghai-Tibet Plateau region.Cadmium(Cd)-induced plant senescence can reduce the yield and quality of crops,then bring threat to human health.The purpose of this study was to explore the differences in senescence of two types of highland barley leaves under cadmium stress.Two types of highland barley,’Kunlun12’(late-stage senescing,LS)and’Dulihuang’(early-stage senescing,ES)were used as test materials,after liquid cultured,and then selected to cadmium treatment.The cadmium chloride with concentrations of 25,50,75,100,150μmol/L was selected to treat the highland barley seedlings for 2 days.The cadmium tolerance of two kinds of highland barley was compared by observing their growth and determining the related physiological indices,and the effect of cadmium stress on the senescence process of two kinds of highland barley was explored.The results showed that cadmium treatment caused the two leaves aging of highland barley,which depended on the increase of cadmium concentration,such as yellow leaves,curly tip,decrease of chlorophyll content,increased membrane damage,hydrogen peroxide and cadmium accumulation.Cadmium treatment resulted the increase of CAT activity in the two varieties.The difference was that CAT activity in ES increased first and then decreased with the increase of Cd concentration,while it showed a continuous trend of increase in LS.The activities of the other three antioxidant enzymes(SOD,POD,APX)showed a trend of increasing first and then decreasing with the increase of Cd concentration,and they were significantly higher in ES than in LS.The low concentration cadmium had little effect on LS,while the high concentration of cadmium made both LS and ES exhibit senescence,there were differences in the aging process of the two types of highland barley.When the cadmium concentration reached 75μmol/L,the growth of the two types of highland barley was affected significantly.These results indicated that LS was more tol
