摘要
YABBY基因家族是种子植物特有的转录因子,在植物生长发育过程中起着重要的调控作用。为探究荷花YABBY基因家族的成员和作用,本研究对荷花YABBY基因家族进行了全基因组分析,鉴定到9个YABBY基因,共18个转录本,编码18个蛋白序列。根据系统进化树将其分类为5个亚族,均含有YABBY结构域和C2C2锌指结构域;共线性分析鉴定出4对共线基因。荷花YABBY基因含有顺式作用元件数量多少依次为光响应、激素诱导、逆境胁迫和植物生长发育,仅有NnYABBY3含有胚乳表达相关元件,推测其与莲子的发育有关。进一步对荷花花瓣、瓣化及非瓣化的雄蕊和心皮、荷叶、莲壳等组织的转录组数据分析发现,NnYABBY5、NnYABBY6和NnYABBY93个基因在心皮中特异性表达;NnYABBY3和NnYABBY9在幼嫩叶片中高表达,在成熟叶片中不表达;NnYABBY8在非瓣化雄蕊和非瓣化心皮不表达,而在花瓣、瓣化雄蕊和瓣化心皮中均有表达,推测NnYABBY8可能与雄蕊和心皮瓣化有关;NnYABBY3在心皮和莲壳中的表达量显著高于其他组织,推测NnYABBY3与莲子的发育有关。这些结果表明荷花YABBY基因具有调控荷花生长发育的潜在应用价值,为进一步研究荷花YABBY基因功能提供参考和理论基础。
The YABBY gene family is one of the transcription factor families only found in spermatophyte.It plays an essential role in the regulation of growth and development of plants.To analyze the members and functions of the lotus YABBY gene family.In this study,a genome-wide analysis of YABBY gene family in louts was carried out,and 9 YABBY genes were identified,with a total of 18 transcripts,encoding 18 protein sequences.According to the phylogenetic tree,these genes are classified into 5 sub-families,all of which contain YABBY domain and C2C2zinc finger domain.The collinearity analysis identified 4 pairs of collinearity genes.The number of cis-acting elements contained in lotus YABBY gene is in order of light response,hormone induction,adversity stress and plant growth and development,only NnYABBY3 contains endosperm expression-related elements,which is assumed to be related to the development of lotus seeds.Further analysis of the transcriptome data of the lotus petals,petalized and non-petalized stamens and carpels,lotus leaves,lotus shells and other tissues found that the 3 genes NnYABBY5,NnYABBY6 and NnYABBY9 express specifically in the carpel.NnYABBY3 and NnYABBY9 express highly in young leaves but do not express in mature leaves.NnYABBY8 does not express in non-petalized stamens and non-petalized carpels,but express in petals,petalized stamens and petalized carpels,indicating that NnYABBY8may relate to petalization of stamens and carpels.The expression of NnYABBY3 in carpel and lotus shell is significantly higher than in other tissues.This is an indication that NnYABBY3 is related to the development of lotus seeds.These results indicate that the lotus YABBY genes potentially regulate the growth and development of the lotus,which will be a reference for further research on the function of the lotus YABBY genes.
