川芎不忘散对阿尔茨海默病模型小鼠学习记忆能力及海马组织血管内皮生长因子表达的影响

摘要 目的:探究川芎不忘散对阿尔茨海默病(Alzheimer's Disease,AD)模型小鼠学习记忆能力及海马组织血管内皮生长因子(Vascular Endothelial Growth Factor,VEGF)表达的影响。方法:选取8周龄无特定病原体级PP/PS1双转基因雄性小鼠24只作为AD模型动物,正常小鼠6只为正常组,将24只AD小鼠分为安慰组、低剂量组、常规剂量组、高剂量组四个组,每组6只。应用Morris水迷宫试验和旷场实验连续观察各组AD小鼠在第1周、第2周、第3周、第4周的认知功能变化。通过实时荧光定量聚合酶链式反应(Real-time Fluorescence Quantitative Polymerase Chain Reaction,RT-qPCR)和蛋白免疫印迹法检测小鼠海马组织中VEGF蛋白的表达水平。结果:Morris水迷宫试验和旷场实验结果显示,与安慰组相比,低剂量组、常规剂量组、高剂量组小鼠在治疗1、2、3、4周后逃避潜伏期时间均逐渐缩短,运动总距离逐渐增加(P<0.05),呈剂量效应变化(P<0.05),但仍明显高于或低于正常组(P<0.05)。RT-qPCR和蛋白免疫印迹法结果显示与安慰组相比,低剂量组、常规剂量组、高剂量组小鼠VEGF mRNA和蛋白表达均逐渐增加(P<0.05),且呈剂量效应增加(P<0.05),但仍明显低于正常组(P<0.05)。结论:高剂量川芎不忘散AD模型小鼠的学习记忆能力提高,其作用机制可能与海马组织VEGF较高的表达水平有关。 Objective:To investigate the effects of Chuanxiong Buwang San(川芎不忘散)on learning and memory ability and expression of VEGF in hippocampus of AD model mice.Methods:A total of 24 eight-week-old specified pathogen free grade PP/PS1 double transgenic male mice were selected as AD model animals,and 6 normal mice were included in the normal group.The 24 AD model animals were divided into four groups:the comfort group,the low dose group,the conventional dose group and the high dose group,with 6 model animals in each group.Morris water maze test and open field test were used to observe the changes of cognitive function of AD mice in each group at 1,2,3 and 4 weeks.The expression of VEGF protein in hippocampus of model mice was detected by RT-qPCR and western blot.Results:The results of Morris water maze test and open field test showed that compared with the comfort group,the escape latency time of model mice in the low dose group,the conventional dose group and the high dose group of Chuanxiong Buwang San was gradually shortened after 1,2,3 and 4 weeks of treatment,and the total movement distance was gradually increased(P<0.05),with a dose-effect change(P<0.05).However,the two indicator above were still significantly higher than or lower than the normal group(P<0.05).The results of RT-q PCR and western blot showed that compared with the comfort group,VEGF mRNA and protein expressions in the low dose group,the conventional dose group and the high dose group of Chuanxiong Buwang San were gradually increased(P<0.05),with a dose-effect change(P<0.05),but still significantly lower than the normal group(P<0.05).Conclusion:High-dose Chuanxiong Buwang San can improve the learning and memory ability of AD model mice,and its action mechanism may be related to the higher expression of VEGF in hippocampus.
