摘要
目的探讨食醋调吴茱萸穴位贴敷对对氯苯丙氨酸(PCPA)所致失眠大鼠在体皮肤渗透过程及作用机制。方法SPF级7周龄SD雄性健康大鼠162只分成正常组,模型组,阳性对照组(地西泮1 mg/kg,10 ml/kg),食醋调吴茱萸小、中、大剂量组(剂量为2、4、8 g/kg;每组分为涌泉穴组和神阙穴组)。正常组、模型组和阳性对照组(地西泮)每组10只,其余各组22只,其中10只用于观察脑组织神经递质γ-氨基丁酸(GABA)含量及GABAA1 mRNA表达,另外12只用于观察食醋调吴茱萸穴位贴敷对大鼠在体皮肤的渗透过程。除正常组外其余各组均使用PCPA法造模。给药组每天贴敷给药4 h,连续给药7 d后,获取大鼠脑组织标本,采用酶联免疫吸附试验法检测神经递质GABA含量,采用实时荧光定量PCR检测GABAA1 mRNA的表达水平。第7天给药结束后1 h(T_(1))、给药结束后4 h(T_(2))、给药结束后8 h(T_(3)),以及贴敷给药8 h清除药物后1 h(T_(4)),取大鼠涌泉穴、神阙穴部位皮肤制作冰冻切片,用激光共聚焦显微镜观察食醋调吴茱萸穴位贴敷的在体皮肤渗透过程。结果与模型组比较,食醋调吴茱萸穴位贴敷大、中、小剂量组大鼠下丘脑、脑干、海马GABA含量升高,差异有统计学意义(P<0.05);食醋调吴茱萸穴位贴敷中、高剂量组大鼠下丘脑、脑干、海马GABAA1 mRNA表达升高,差异有统计学意义(P<0.05)。两个穴位食醋调吴茱萸穴位贴敷大、中、小剂量组组内不同时间点平均荧光强度比较,差异有统计学意义(P<0.05);两个穴位食醋调吴茱萸穴位贴敷大、中、小剂量组T_(1)~T_(4)时平均荧光强度比较,差异有统计学意义(P<0.05)。结论食醋调吴茱萸穴位贴敷具有改善失眠的作用,其作用机制可能与上调脑组织GABA含量及GABAA1 mRNA的表达水平有关。食醋调吴茱萸穴位贴敷具有良好的经皮渗透性。
Objective To explore skin penetration process and mechanism of acupoint application of evodia rutaecarpa mixed with vinegar in the treatment of para-chlorophenylalanine(PACA)insomnia rats.Methods One hundred and sixty-two SPF grade 7-week-old healthy SD male rats were divided into normal group,model group,positive control group(Diazepam 1 mg/kg,10 ml/kg),small,medium,and large dose groups(doses of 2,4,8 g/kg;each group was divided into yǒngquán point group and shénquēpoint group).There were 10 rats in normal group,model group,and positive control group(Diazepam),and 22 rats in other groups.Ten rats were used to observe the content of the neurotransmitterγ-aminobutyric acid(GABA)in brain tissue and the expression of GABAA1 mRNA,and the other 12 rats were used to observe the penetration process of the acupoint application of evodia rutaecarpa mixed with vinegar in body skin.Except normal group,the other groups were modeled by PCPA method.The rats in the administration group were affixed and administered for 4 h a day,and after 7 d of continuous administration,brain tissue samples were obtained,and the content of the neurotransmitter GABA was detected by enzyme linked immunosorbent assay,and the expression level of GABAA1 mRNA was detected by real-time PCR.After 7 days to 1 h(T_(1)),4 h after drug(T_(2)),after 8 h(T_(3)),and 8 h after application and 1 h after removal of the drug(T_(4)),the skin of yǒngquán point and shénquēpoint of rats was frozen and sliced,the penetration process of acupoint application of evodia rutaecarpa mixed with vinegar was observed by confocal laser scanning micrography.Results Compared with model group,GABA content in hypothalamus,brainstem,and hippocampus of rats in large,medium and small dose groups of acupoint application of evodia rutaecarpa mixed with vinegar were increased,and the differences were statistically significant(P<0.05);GABAA1 mRNA expression of hypothalamus,brainstem,and hippocampus of rats in middle and high dose groups of acupoint application of evodia rutaeca
