摘要
目的探索补肾健脾活血方调控Wnt/β-连环蛋白(β-catenin)通路对复发性流产(RSA)小鼠母胎界面血管生成的影响。方法将CBA/J雌性小鼠与DBA/2雄性小鼠合笼交配建立RSA小鼠模型,并随机分为模型组、补肾健脾活血方组、KYA1797K组、补肾健脾活血方+KYA1797K组,每组12只;将CBA/J雌性小鼠与BALB/c雄性小鼠合笼交配所得的12只正常妊娠小鼠作为对照组。计算各组小鼠胚胎丢失率;检测各组小鼠蜕膜组织病理形态、血小板-内皮细胞黏附分子(CD31)表达情况;检测各组小鼠血清雌二醇(E_(2))、孕酮(P)、血管内皮生长因子(VEGF)含量及蜕膜组织Wnt/β-catenin通路蛋白表达情况。结果与对照组相比,模型组小鼠蜕膜组织呈明显病理损伤,CD31阳性细胞比例、Wnt1及β-catenin蛋白表达量明显减少,血清中E、P及VEGF含量明显减少,胚胎丢失率明显增加(P<0.05)。与模型组、补肾健脾活血方+KYA1797K组相比,补肾健脾活血方组小鼠蜕膜组织病理损伤减轻,CD31阳性细胞比例、Wnt1及β-catenin蛋白表达量增加,血清中E_(2)、P及VEGF含量减少,胚胎丢失率减少(P<0.05)。结论补肾健脾活血方可激活Wnt/β-catenin通路,进而提高雌激素和孕激素水平,促使RSA小鼠母胎界面血管生成,从而提高保胎成功率。
Objective To explore the impact of Bushen Jianpi Huoxue Recipe on the angiogenesis at the maternal-fetal interface of recurrent spontaneous abortion(RSA)mice through Wnt/β-catenin pathway.Methods CBA/J female mice and DBA/2 male mice were mated in cages to establish RSA mouse model.The model was randomly grouped into model group,Bushen Jianpi Huoxue Recipe group,KYA1797 K group,and Bushen Jianpi Huoxue Recipe+KYA1797 K group,12 per group.CBA/J female mice and BALB/c male mice were mated in cages to obtain 12 normal pregnant mice,which were set as the control group.The embryo loss rate of mice in each group was calculated;the pathological morphology of decidua,the expression of platelet-endothelial cell adhesion molecule(CD31)were detected;and the contents of estradiol(E_(2)),progesterone(P)and vascular endothelial growth factor(VEGF)in serum and the expression of Wnt/β-catenin pathway protein in decidua were detected.Results Compared with the control group,the decidua tissue of the model group showed obvious pathological damage,the proportion of CD31 positive cells and the expression of Wnt1 andβ-catenin proteins were obviously reduced,the contents of E,P and VEGF in serum were obviously reduced,and the embryo loss rate was obviously(P<0.05).Compared with the model group and the Bushen Jianpi Huoxue Recipe+KYA1797 K group,the decidual tissue pathological damage of mice in the Bushen Jianpi Huoxue Recipe group was reduced,the proportion of CD31 positive cells and the expression of Wnt1 and β-catenin proteins were increased,the contents of E_(2),P and VEGF in serum were obviously reduced,and the embryo loss rate was obviously reduced(P<0.05).Conclusions Bushen Jianpi Huoxue Recipe can activate Wnt/β-catenin pathway to increase the levels of estrogen and progesterone,promote angiogenesis at the maternal fetal interface of RSA mice,and improve the success rate of fetal protection.
