摘要
目的观察蒙药德都红花-7味散对四氯化碳所致肝纤维化大鼠肝组织铁蓄积及CDGSH铁硫结构域(CISD1)、谷胱甘肽过氧化酶4(GPX4)、二价金属转运蛋白1(DMT1)、线粒体铁蛋白(FTMT)表达的影响。方法将40只SD雄性大鼠按体质量分层随机分为空白组、模型组、对照组和实验组,每组10只。采用腹腔注射CCl_(4)方法建立肝纤维化模型,空白组和模型组灌胃等量蒸馏水,实验组灌胃德都红花-7味散(0.3 g·kg^(-1)),对照组灌胃水飞蓟宾(21 mg·kg^(-1)),每天给药1次,给药7周后动物禁食12 h,取肝组织4%多聚甲醛固定。普鲁士蓝染色观察肝组织内铁蓄积,反转录聚合酶链反应(RT-PCR)方法检测大鼠肝组织中CISD1、GPX4、DMT1、FTMT mRNA的表达。结果空白组、模型组、对照组和实验组普鲁士蓝染色面积百分比分别为0,(1.37±0.48)%,(0.31±0.11)%和(0.68±0.16)%,与空白组比较模型组、对照组、实验组普鲁士蓝染色面积百分比升高,差异有统计学意义(P<0.05)。与模型组比较,对照组、实验组普鲁士蓝染色面积百分比均下降,差异均有统计学意义(均P<0.05)。空白组、模型组、对照组、实验组CISD1 mRNA表达分别为1.07±0.15,1.51±0.43,1.06±0.18,0.84±0.37;GPX4 mRNA表达分别为2.09±0.57,0.99±0.16,1.26±0.31,1.32±0.37;DMT1 mRNA表达分别为0.62±0.17,0.83±0.17,0.43±0.08,0.43±0.14;FTMT mRNA表达分别为1.12±0.24,0.76±0.20,0.79±0.14,1.10±0.23,与空白组比较,模型组CISD1、DMT1 mRNA表达升高,GPX4、FTMT mRNA表达降低,差异均有统计学意义(均P<0.05);与模型组比较,对照组、实验组CISD1、DMT1 mRNA表达下降,对照组GPX4 mRNA、实验组GPX4、FTMT mRNA表达升高,差异均有统计学意义(均P<0.05)。结论蒙药德都红花-7味散能够明显减轻CCl_(4)所致慢性肝损伤,其机制与减少肝组织内铁蓄积、调节CISD1、GPX4、DMT1、FTMT mRNA表达相关。
Objective To observe the effects of Mongolian medicine Deduhonghua-7 powder on the expression of CDGSH iron sulfur domain1(CISD1),glutathione peroxidase 4(GPX4),divalent metal transportor1(DMT1)and mitochondrial ferritin(FTMT)in liver tissue of rats with CCl_(4)-induced liver fibrosis.Methods Forty SD male rats were equally divided into blank group,model group,control group and experimental group at random with 10 rats in each group.We established the model of hepatic fibrosis through intraperitoneal injection of CCl4.The blank group and the model group wereprovided with distilled water,the experimental group were offered Deduhonghua-7 powder(0.3 g·kg^(-1)),and thecontrol group were given silymarin(21 mg·kg^(-1))once a day.The animals were fasted for 12 h after seven weeks’administration,and their liver tissue was fixed by 4%paraformaldehyde.The iron accumulation in liver tissue wasobserved by Prussian blue staining,and the mRNA expressions ofCISD1,GPX4,DMT1 andFTMTin rat liver tissuewere detected by reverse transcription polymerase chain reaction(RT-PCR).Results The percentage of Prussianblue staining area in the blank,model,control and experimental groups are 0,(1.37±0.48)%,(0.31±0.11)%and(0.68±0.16)%respectively.In comparison to the blank group,the percentage of Prussian blue staining area inthe model group,control group and the experimental group increased significantly(allP<0.05).In comparison to themodel group,the percentage of Prussian blue staining area in the control group and the experimental group decreasedsignificantly(P<0.05).CISD1 mRNA expressions in the blank,model,control,and experimental groups were1.07±0.15,1.51±0.43,1.06±0.18 and 0.84±0.37 respectively;GPX4 mRNA expressions were 2.09±0.57,0.99±0.16,1.26±0.31 and 1.32±0.37 respectively;DMT1 mRNA expressions were 0.62±0.17,0.83±0.17,0.43±0.08 and 0.43±0.14 respectively,and FTMTmRNA expressions were 1.12±0.24,0.76±0.20,0.79±0.14 and 1.10±0.23 respectively.In comparison to the blank group,the expression ofCISD1 andDMT1mRNA in
