miR-214在绵羊毛囊周期性发育中的调控作用研究 被引量:1

Regulatory effects of miR-214 on the periodic development of sheep hair follicles

摘要 为了探讨miR-214在绵羊毛囊生长发育过程中的调控作用,试验分析了来源于前期转录组测序技术获得的差异性表达的miR-214序列,并采用生物信息学方法预测靶基因,结合GO功能注释和KEGG富集分析对预测的靶基因进行研究。结果表明:miR-214在各物种中高度保守,经在线软件YM500V2预测miR-214主要在皮肤组织高表达,且miR-214在毛囊发育的生长期低表达,休止期高表达。miR-214共靶向到1045个靶基因,其中26个位于3条毛囊发育相关信号通路WNT、P53、FOXO中。PCK2、CCNB3、SKP2、RRM2、SFN和CCNE2基因的相关系数低于-0.75,且各有3个位于P53和FOXO3信号通路。上述6个靶基因随着毛囊生长阶段的发展呈现规律变化,在生长期高表达,休止期低表达,生长早期再次高表达。说明miR-214可能通过在休止期的高表达导致FOXO、P53、WNT信号通路上靶基因的低表达,以起到重要的调控作用。 To explore the regulatory role of miR-214 during hair follicle growth and development in sheep, the differentially expressed miR-214 sequences obtained by transcriptome sequencing were analyzed. Bioinformatics methods were used to predict miRNA target genes, and GO functional annotation and KEGG enrichment analysis were used to study the predicted target genes. The results showed that miR-214 was highly conserved among species. The online software YM500 V2 predicted that miR-214 was mainly highly expressed in skin tissue, and miR-214 was low in the growth phase and high in the resting phase of hair follicle development. A total of 1 045 target genes were targeted by miR-214, among which 30 were located in three hair follicle development related signaling pathways, WNT, P53 and FOXO. The correlation coefficients of PCK2, CCNB3, SKP2, RRM2, SFN and CCNE2 genes were lower than-0.75, and three of them were located in the P53 and FOXO3 signaling pathways. The six target genes changed regularly with the development of hair follicle growth stages, with high expression in the anagen phase, low expression in the telogen phase, and high expression again in the anagen phase. It indicated that miR-214 contributed to the low expression of target genes on FOXO, p53 and WNT pathways through the high expression in telogen, which played an important regulatory role in hair follicle telogen.
