杜泊羊LEF1生物信息学及表达分析

摘要 旨在对杜泊羊LEF1基因进行生物信息学分析和编码蛋白功能特征预测,探究其对毛囊发育的调控机制,及其在毛囊不同发育时期中的表达量。从试验羊场选择2周岁体重相近的5只杜泊母羊,利用生物信息学方法比对绵羊LEF1氨基酸序列与其他物种的亲缘性,分析LEF1蛋白质的理化性质和结构特征;利用实时荧光定量PCR检测其在杜泊羊体测腹部皮肤的表达量。结果表明,杜泊羊LEF1蛋白的氨基酸序列与牛的亲缘关系最近,与小鼠的亲缘关系最远。LEF1基因编码的氨基酸331个;LEF1基因编码的蛋白质分子式C_(1636)H_(2534)N_(478)O_(481)S_(15),相对分子质量37075.95 ku,理论等电点数值9.52,亲水性平均值-0.906;编码蛋白的二、三级结构主要由无规卷曲(57.7%)和α-螺旋(28.1%)组成,同时包含1个糖基化位点。LEF1基因在杜泊羊毛囊不同发育时期均有表达,在9月和3月高表达,在1月低表达。综上,LEF1基因在杜泊羊毛囊的生长期高表达。 Aimed to bioinformatic analysis and prediction of functional characteristics of the encoded protein on LEF1 gene in Dorper sheep,to explore its regulatory mechanism on hair follicle development,and its expression quantity in different developmental stages of hair follicle.Five Dorper ewes of similar body weight at 2 weeks of age were selected from the testing goat farm,and the amino acid sequence of ovine LEF1 was aligned with other species using bioinformatics methods to analyze the physicochemical properties and structural characteristics of LEF1 protein;real time fluorescence was used to quantify PCR expression in the somatometric abdominal skin of Dorper sheep.The results showed that the ovine LEF1 amino acid sequence was the most closely related to bovine and least related to mouse.331 amino acids were encoded by LEF1;the protein encoded by the LEF1 gene had a molecular formula of C_(1636)H_(2534)N_(478)O_(481)S_(15),a relative molecular mass was 37075.95 Ku,a theoretical isoelectric point value was 9.52,and an average value of hydrophilicity was-0.906;the secondary tertiary structure of the encoded protein was dominated by random coils(57.7%)andα-Helix(28.1%)composition while containing 1 glycosylation site.The LEF1 gene was expressed in different developmental stages of the hair follicle in Dorper sheep,with high expression in September and March,and low expression in January.In conclusion,the high expression of LEF1 gene during the anagen period of Dorper sheep.
