摘要
目的分析归元剔络养血方(GYTL)对非动脉炎性前部缺血性视神经病变(NAION)模型大鼠血管内皮生长因子(VEGF)的调控作用。方法SD雄性大鼠36只,随机分为正常组,模型组和中药组3组,每组12只。正常组常规饲养;模型组和中药组在采用光动力法制备NAION大鼠模型成功后,分别给予蒸馏水3 mL和中药GYTL 30 g/kg每日灌胃1次,于第10 d取出大鼠视网膜,苏木素-伊红(HE)染色观察大鼠视网膜病理变化;免疫组织化学法测定视网膜VEGF表达,并进行平均光密度检测;酶联免疫吸附测定法(ELISA)测定玻璃体及外周血VEGF含量。结果(1)组织病理学:与正常组比较,模型组、中药组存在不同程度视网膜神经纤维层(RNFL)水肿和视神经节细胞(RGCs)凋亡,而中药组RNFL水肿、RGCs凋亡较模型组轻。组间视网膜VEGF平均光密度差异有统计学意义(F=19.851,P=0.004);模型组视网膜VEGF平均光密度较正常组升高(t=6.299,P=0.001);中药组低于模型组(t=-2.966,P=0.031),高于正常组(t=3.646,P=0.015),均有统计学意义。(2)玻璃体及外周血中VEGF含量:组间血清VEGF浓度比较有统计学意义(F=5.440,P=0.018);模型组血清VEGF浓度较正常组升高(t=2.803,P=0.014);中药组较模型组降低(t=-2.787,P=0.015),均有统计学意义。而3组间玻璃体VEGF浓度差异无统计学意义(P>0.05)。结论在NAION早期,GYTL可以抑制视网膜VEGF过度表达,这可能是其减轻病理反应,保护RGCs的生物学基础之一。
OBJECTIVE To analyze the regulatory effect of Guiyuan Tiluo Yangxue Formula(GYTL)on vascular endothelial growth factor(VEGF)in the rats with non-anteritic amterior ischemic optic neuropathy(NAION).METHODS A total of 36 male SD rats were randomly divided into normal group,model group and Traditional Chinese Medicine(TCM)group,with 12 rats in each group.The rats of normal group were fed routinely,whereas the rats of model group and TCM group were fed with 3 mL of distilled water and 30 g/kg of GYTL by gavage once a day respectively after the successful establishment of NAION rat model by photodynamic method.The retinas of the rats were obtained on the 10^(th) day,and hematoxylin-eosin staining(HE)was performed to observe the pathological changes of the retina.The expression of VEGF in retinas was detected by immunohistochemistry,and the average optical density(AOD)was measured.The expression level of VEGF in vitreous and peripheral blood were detected by enzyme-linked immunosorbent assay(ELISA).RESULTS(1)Histopathology:Compared with the normal group,the edema of retinal nerve fiber layer(RNFL)and the apoptosis of retinal ganglion cells(RGCs)were present in model group and TCM group;and RNFL edema and RGCs apoptosis in TCM group were lighter than those in model group.There was statistical difference in AOD of retinal VEGF between three groups(F=19.851,P=0.004);AOD of retinal VEGF in model group was higher than that in normal group(t=6.299,P=0.001);AOD of retinal VEGF in TCM group was lower than that in model group(t=-2.966,P=0.031),and higher than that in normal group(t=3.646,P=0.015),all with statistical significance.(2)VEGF expression level in vitreous and peripheral blood:There was statistical difference in serum VEGF expression level between three groups(F=5.440,P=0.018).The serum VEGF expression in model group was higher than that in normal group(t=2.803,P=0.014);The serum VEGF expression in TCM group was lower than that in model group(t=-2.787,P=0.015).However,there was no statistical difference in vitreous VE
