小檗碱对被动吸烟大鼠血清血管炎性因子影响的研究

摘要 目的:观察被动吸烟对大鼠血清超敏C反应蛋白(hs-CRP)、白细胞介素-6(IL-6)、基质金属蛋白酶-9(MMP-9)等血管炎性因子的影响。观察灌服小檗碱溶液对被动吸烟大鼠血清hs-CRP、IL-6、MMP-9的影响。方法:实验一:按吸烟持续时间不同将实验大鼠随机均分为吸烟长时间组(n=3)、吸烟短时间组(n=3)、非吸烟对照组(n=3),观察持续不同时间的被动吸烟对大鼠血清中hs-CRP、IL-6、MMP-9等血管炎性因子的影响。实验二:将实验大鼠随机均分为吸烟长时间组(n=9)和吸烟长时间灌药组(n=9),吸烟长时间灌药组每天给大鼠予小檗碱溶液按60mg/(kg·d)的剂量进行灌胃,分析小檗碱对相同持续时间的被动吸烟大鼠血清中hs-CRP、IL-6、MMP-9的影响。结果:实验一:hs-CRP含量在吸烟长时间组中的含量低于非吸烟对照组和吸烟短时间组(P<0.05),但在非吸烟对照组和吸烟短时间组之间无差异(P>0.05);IL-6含量在吸烟长时间组中的含量高于非吸烟对照组和吸烟短时间组(P<0.05),但在非吸烟对照组和吸烟短时间组之间无差异(P>0.05)。MMP-9含量在吸烟长时间组中的含量高于非吸烟对照组和吸烟短时间组(P<0.05),但在非吸烟对照组和吸烟短时间组之间无差异(P>0.05)。实验二:与吸烟长时间组相比,hs-CRP含量在吸烟长时间灌药组中无显著差异(P>0.05),但IL-6和MMP-9含量在吸烟长时间灌药组中均有降低,有显著差异(P<0.05)。结论:被动吸烟能够时间依赖地使大鼠血清IL-6、MMP-9等血管炎性因子水平升高,提示这可能是吸烟导致动脉粥样硬化的具体病理学机制之一。灌服小檗碱溶液可以抑制被动吸烟大鼠血清IL-6、MMP-9水平的升高,从而可以起到预防和治疗动脉粥样硬化的作用。 Objective:To observe the effects of passive smoking on serum hypersensitive C-reactive protein(hs-CRP),interleukin-6(IL-6)and matrix metalloproteinase-9(MMP-9)of rats.To observe the effects of berberine solution on serum hs-CRP,IL-6 and MMP-9 in rats exposed to passive smoking.Methods:Experiment 1:According to the different duration of smoking,the rats were randomly divided into three groups:long-term smoking group(n=3),short-term smoking group(n=3)and non-smoking control group(n=3),and the effects of different duration of passive smoking on serum hs-CRP,IL-6,MMP-9 and other vascular inflammatory factors were observed.Experiment 2:The rats were randomly divided into the long-term smoking group(n=9)and the long-term smoking and drug infusion group(n=9).The berberine solution was given to the rats by gavage at a dose of 60mg/(kg·d)every day,and the effects of berberine on the serum hs-CRP,IL-6 and MMP-9 of the passive smoking rats with the same duration were analyzed.Results:Experiment 1:The hs-CRP content in the long-term smoking group was lower than that in the non-smoking control group and the short-term smoking group(P<0.05),but there was no difference between the non-smoking control group and the short-term smoking group(P>0.05).IL-6 levels were higher in the long-term smokers than in the non-smoking control group and the short-term smokers(P<0.05),but there was no difference between the non-smoking control group and the short-term smokers(P>0.05).The content of MMP-9 in the long-term smoking group was higher than that in the non-smoking control group and the short-term smoking group(P<0.05),but there was no difference between the non-smoking control group and the short-term smoking group(P>0.05).Experiment 2:There was no significant difference in hs-CRP content in the long-term smoking group compared with the long-term smoking and drug infusion group(P>0.05),but the IL-6 and MMP-9 contents were decreased in the long-term smoking and drug infusion group,showing a significant difference(P<0.05).Conclusion:Pass
