百合AP2/ERF家族转录因子基因LoERF4的克隆和生物信息学分析 被引量:1

摘要 【目的】研究AP2/ERF家族成员ERF基因在百合休眠解除过程中的作用。【方法】以百合品种西伯利亚(Lilium oriental‘Siberia’)的小鳞茎为试验材料,从前期试验获得的转录组测序数据中筛选得到1个在小鳞茎休眠解除过程中显著上调表达的ERF转录因子,结合RT-PCR技术扩增得到西伯利亚小鳞茎的ERF基因cDNA全长序列,命名为LoERF4;通过多种生物信息学软件对LoERF4的基因结构和理化性质等进行分析;利用荧光定量qRT-PCR技术检测LoERF4基因在小鳞茎休眠解除过程中的相对表达量。【结果】通过克隆得到LoERF4基因全长,该基因开放阅读框全长为606 bp,编码201个氨基酸,含有1个典型的保守AP2结构域;蛋白相对分子量约为21.52 ku,理论等电点为6.43,为亲水性蛋白,没有跨膜结构。qRT-PCR分析结果显示:在西伯利亚小鳞茎休眠解除过程中,该基因的表达量在第20天小鳞茎休眠解除、开始萌发时极显著升高。【结论】在百合中克隆得到的LoERF基因可能是参与调控百合小鳞茎休眠解除进程的关键因子之一。 [Purpose]To study the role of ERF genes,members of the AP2/ERF family,in the dormancy release process of lily.[Methods]Bulblets of Lilium oriental‘Siberia’were used as experimental materials.From the transcriptome sequencing data obtained in the previous experiments,we screened for an ERF transcription factor that was significantly up-regulated in the dormancy release process of bulblets.The full-length cDNA sequence of ERF gene of bulblets of L.oriental‘Siberia’,named LoERF4,was obtained by combining RT-PCR.The LoERF4 gene structure and physicochemical properties were bioinformatically analyzed by various bioinformatics software.The relative expression level of LoERF4 gene during dormancy release in bulblets was examined by qRT-PCR.[Results]The full length of LoERF4 gene was obtained by cloning,which had a full open reading frame with length of 606 bp,encoded 201 amino acids and contained a typical conserved AP2 structural domain.The relative molecular weight of the protein was about 21.52 ku,and the theoretic-al isoelectric point was 6.43.It was a hydrophobic protein without transmembrane structure.qRT-PCR analysis result showed that the expression of this gene extremely significantly increased at the beginning of dormancy release of bulblets of L.oriental‘Siberia’on the 20th day.[Conclusion]The LoERF gene is cloned in lily,and it is hypothesized that the LoERF gene may be one of the key factors involved in regulating the process of dormancy release in lily bulblets.
